Is dark tea related to CYP450(cytochromeP450)metabolism? So far,no literature has been reported.The experiment to tea in the important monomer EGCG and all kinds of dark tea(Fu brick tea,Bailiang tea,Raw dark greentea and bulk dark green tea for fermentation)as the experimental materials,C57 BL /6 female mice as the research material,by q-PCR and Western blot and other experimental methods.The effects of EGCG and various dark tea on the relative expression of PXR and CYP450 m RNA and the expression of PXR and AhR protein in the viscera of mice were studied.Results showed that:1.C57BL/6 mice generally showed an increase in the intake of mice and an increase in weight gain in terms of food intake and body weight.2.The relative expression of PXR and CYP450 m RNA in the colon of mice showed: The treatment groups compared with the blank group,EGCG up-regulates the expression of PXR gene;The expression of PXR and CYP3A9 genes was up-regulated by Raw dark green tea;The expression of PXR,CYP1A2,CYP2B1 and CYP3A1 genes was up-regulated by Fu brick tea;The expression of CYP1A2?CYP2B1?CYP3A1and CYP3A9 genes was up-regulated by Bailiang tea;The expression of PXR genes was up-regulated by Bulk dark green tea for fermentation.3.The relative expression of PXR and CYP450 m RNA in mice liver showed: The treatment groups compared with the blank control group,EGCG up-regulates the expression of CYP1A2?CYP2C6 and CYP3A1 gene;The expression of PXR?CYP1A2?CYP2C6 and CYP3A9 genes was up-regulated by Raw dark green tea;The expression of CYP1A2 and CYP2C6 genes was up-regulated by Fu brick tea;The expression of CYP1A2?CYP2C6 and CYP3A9 genes was up-regulated by Bailiang tea;The expression of PXR?CYP1A2?CYP2B1?CYP2C6?CYP3A1 and CYP3A9 genes was up-regulated by Bulk dark green tea for fermentation.4.The changes of PXR and AhR protein expression in the colon and liver of mice were as follows: EGCG,Raw dark green tea,Fu brick tea and Bailiang tea significantly increased the relative expression of PXR protein in the colon of mice,while The Bulk dark green tea for fermentation was no signif icant effect.AhR is basically not expressed in the colon.The relative expression of PXR protein in mice liver was significantly increased byThe Bulk dark green tea for fermentation,while EGCG,Raw dark green tea,Fu brick tea and Bailiang tea slightly reduced the relative expression of PXR protein.Raw dark green tea,Fu brick tea and Bailiang tea were signif icantly increased the relative expression of AhR protein in mice liver,while EGCG had no obvious effect.Therefore,EGCG and all kinds of dark tea feeding mice will affect the expression of PXR and CYP450-related genes in mice,and the amount of PXR and CYP1A2 could signif icantly increase the expression of PXR and CYP1A2,as well as enhance the expression of PXR and AhR protein Expression,other types of dark tea also have a certain role in upgrading.So dark tea,especially the Bulk dark green tea for fermentation a through the impact of CYP450,which reflects a certain pharmacological health care function.
