Stroke is one of the major diseases that seriously threaten human health.Natural antistroke drugs have definite curative effect and little side effect.However,the development of natural drugs has been restricted due to its complex composition and unclear mechanism of action.Therefore,it is of great significance to separate and evaluate the chemical constituents and activity of natural drugs.In this paper,the isoflavones in Belamcanda chinensis of Chinese medicine are systematically analyzed.Firstly,the extraction technology of isoflavones in Belamcanda chinensis with traditional organic solvents and ultrasonic assisted ionic liquid was optimized.Then using high performance liquid chromatography(HPLC)and mass spectrometry(LC-MS)techniques to analysis the structure of the chemical composition in the extraction of Belamcanda chinensis.,using PC12 cells and ultrafiltration mass spectrometry(UF-LC-MS)activity screening technology to screen the antistroke active compounds in the extraction of Belamcanda chinensis,finally using a type of semi-preparation of high performance liquid chromatography(Semi-prep.HPLC)to separation of active monomers and test the purity by high performance liquid chromatography(HPLC).Four kinds of similar Tectorigenin compounds were extracted and separated from Belamcanda chinensis by ionic liquid-ultrasonic assisted extraction.Seven monomer compounds were isolated by Semi-prep.HPLC,and the purity was greater than 91%.Five lactic acid dehydrogenase inhibitors were screened from traditional organic solvent extracts by PC12 cells and UF-MS.In the experiments,MPP~+damaged cells and extraction of Belamcanda chinensis were mixed culture,the cell survival rate is significantly higher than normal control group,show that cell less damage and protected,cell survival rate increases,the experimental results prove that the extraction of Belamcanda chinensis do have nerve protective effect for the MPP~+damaged PC12 cells.On this basis,the use of ultrafiltration and mass spectrometry method to screen the inhibitior of Lactate dehydrogenase in the extraction of Belamcanda chinensis.With HPLC and electrospray multi-stage tandem mass spectrometry(HPLC-ESI/MS~n)as the research method,to analysis the structure of the active chemical composition in the extraction of Belamcanda chinensis.Four kinds of?-glycosidase inhibitors were screened from ionic liquid-ultrasonic assisted extraction by UF-MS.Topic selection has high application value in the development of new drugs and the treatment of diseases,and has certain significance in ensuring human health and promoting the improvement of people's living standards.Paper has established the isoflavone compounds in Belamcanda chinensis traditional organic solution and ultrasonic assisted ionic liquid extraction method,UF-MS and PC12 cell activity screening method,HPLC and a type of Semi-prep.HPLC purification and analysis method,and obtained five kinds of lactate dehydrogenase inhibitor and four kinds of?-glycosidase inhibitor,for use of the natural antistroke drugs screening provides an important theoretical basis.
