Nano liquid chromatography is an important microscale separation and analysis technique with the merits of high speed,high performance,low cost and high environmental friendliness.Chromatographic stationary phase is the core part of the nano liquid chromatography system which determines both the separation performance and separation selectivity.Therefore,it is great significance to design and fabricate new chromatographic stationary phases with high resolution and high separation selectivity.The full text is divided into three chapters.The first chapter: In the first part,the high performance liquid chromatography stationary phases were reviewed.In the second part,hybrid monolithic columns were reviewed.In the third part,amino acids and ionic liquids were reviewed.In the last part,the conception of this text was introduced.The second chapter: With 1-vinyl-3-butylimidazolium bromide(VBI)and L-cysteine(Cys)as dual functionalized reagents,polyhedral oligomeric silsesquioxane methacryl substituted(POSS-MA)as the crosslinker,a novel amino acid ionic liquid embedded hybrid monolithic column POSS-VBI-Cys was successfully prepared via a “one-pot” process.The obtained POSS-VBI-Cys monolithic column was characterization via Fourier transform infrared spectrometry,scanning electron microscopy,element analysis,nitrogen adsorption/desorption measurement,respectively.The results showed that the POSS-VBI-Cys hybrid monolithic column was successfully prepared.The third chapter: The chromatographic performance of the POSS-VBI-Cys monolithic columns was further evaluated via capillary high performance liquid chromatography.The developed POSS-VBI-Cys monolithic column demonstrated both reversed-phase liquid chromatography and ion-exchange chromatography retention mechanisms.It was successfully utilized to separate hydrophobic alkylbenzenes,hydrophilic nucleosides and nucleotide bases.It was more suitable for separation of polar compounds and intact proteins.The new POSS-VBI-Cys monolithic column was further used for separation of expressed TARG1 protein sample and good separation performance was achieved.
