As a new type of chromatographic separation medium,the monolithic column has the unique advantages of simple preparation method,high permeability and high separation efficiency.Thus,the development of new monolithic columns is of great significance for the increasing requriment for separation and analysis of complex sample.The full text is divided into three chapters.The first chapter includes four parts.In the first part,the monolithic columns were reviewed.In the second part,the research advancements of polyhedral oligomeric silsesquioxane(POSS)in monolithic columns were reviewed.In the third part,the research progress of ionic liquid modified monolithic column was reviewed.In the last part,the conception of this text was introduced.In the second chapter,with polyhedral oligomeric silsesquioxane methacryl substituted(POSS-MA)as the crosslinker,1-allyl-3-vinylimidazolium bromide(AVI)and D-2-allylglycine hydrochloride(AG)as the dual functionalized reagents,a novel amino acid ionic liquid bridged hybrid monolithic column POSS-AVI-AG was successfully prepared via a “one-pot” process.The POSS-AVI-AG monolithic column was characterization via Fourier transform infrared spectrometry,element analysis,scanning electron microscopy,nitrogen adsorption/desorption measurement and so on.The results showed that the POSS-AVI-AG hybrid monolithic column was successfully prepared.In the third chapter,the chromatographic performance of the POSS-AVI-AG monolithic columns was evaluated via capillary high performance liquid chromatography.The POSS-AVI-AG monolithic column possesses typical dual retention mechanism of RPLC and HILIC.POSS-AVI-AG monolithic column showed high column efficiency and excellent separation selectivity in the separation of hydrophobic alkylbenzenes,anilines,phenols and hydrophilic thioureas,nucleosides,nucleics acid bases and amide substances.It was suitable for separation of amide pesticides,cytochrome c digests,standard proteins and egg white proteins with good chromatographic separation performance.The results showed that the POSS-AVI-AG hybrid monolithic column had a certain potential in protein separation thus potential application in proteomics.In the fourth chapter,the summarization and the prospect are presented.
