Sulfonamides and tetracyclines are often used in livestock industry,because of unreasonable used of these antibiotic,plants and animals were affected,as well as the environment were polluted.In this paper,upconversion nanoparticles NaYF4:Yb,Er were prepared by hydrothermal synthesis method and functionalized modification by coated with SiO2 and amino group.And then coupled with sulfonamides and tetracyclines antibodies respectively by the glutaraldehyde cross-linking method which will forming a fluorescence resonance energy transfer system and establishing a method for rapid detection of sulfonamides and tetracyclines veterinary drug residues in animal foods.The main findings are as follows:The less than 100 nm particle sized pure crystal type of UCNPs with high fluorescence yield and good dispersion was synthesized by using the optimized conditions of the molar ratio of sodium citrate/Ln3+is 1:1 and the molar ratio of F-/Ln3+is 6:1 and reaction time of 4 h.Functionalized modification of upconverting nanomaterials was coupled with antibody that will having the 25?g/mL sulfonamides antibody-NaYF4:Yb,Er and 30?g/mL tetracyclines antibody-NaYF4:Yb,Er solution.Meanwhile,the gold nanoparticles synthesized by sodium citrate method,it is less than 10 nm and is spherical in shape.Optimized the concentration of sulfonamides antigen and tetracyclines antigen with gold nanoparticles respectively,then will obtained the solution of 20?g/mL sulfonamides antigen-gold nanoparticles and tetracycline antigen-gold nanoparticles.The fluorescence resonance energy transfer systems of sulfonamides and tetracyclines based on the upconversion nanomaterial-gold nanostructures were constructed,respectively.The sulfonamides antigen detection range was 0.08 to 100ng/mL that is linearly correlated with the recovering of the fluorescence,the lowest detection is 0.08 ng/mL,it also can detect the different types of five-membered heterocycle of sulfonamides antibiotics in milk.The tetracyclines antigen detection ranged from 0.1 to 250 ng/mL,in linear relationship with the recovered fluorescence.The low detection limit is 0.1 ng/mL,it can also be measured in milk and detected different kinds of tetracyclines.In conclusion,upconverting fluorescent nanoparticles and gold nanomaterials are synthesized by optimizing the preparation conditions of nanomaterials.Based on its unique luminescent properties,the fluorescence resonance energy transfer system is formed that can develop fast,simple,high sensitivity and strong specificity assays for the determination of sulfonamides and tetracyclines,respectively.In which provides a good basis and reference for establishing effective detection method.
