Polysaccharide,the most abundant biopolymer in nature,exists in almost all creatures.It is one of the four basic material life activities,which is closely related to the variety of life functions.polysaccharide has many biological functions,including antitumor,antioxidation and so on.It is widely used in various life activities such as cell growth and metabolism,which has little effect on the body.Imperial chrysanthemum,belonging morifolium genus and perennial herb,is one of Chinese traditional herbal medicine and possess extraordinary high edible and medicinal value.This paper mainly studies the extraction,separation,purification and biological structure from Imperial chrysanthemum polysaccharide.On the basis of the above,a chrysanthemum polysaccharide yogurt is developded.The main methods and results are as followed:According to the method of GB and references,the basic components of the chrysanthemum were analyzed,and the results showed that the moisture content was 6.14±0.02%,the total sugar was 23.96 ±0.22%,the protein was 14.88 ±0.35%,the fat was 8.76±0.28%,the raw sugar was 3.40±0.05%,the ash was 8.00±0.15%,and the crude fiber was 7.36±0.05%.The optimal extraction conditions of hot water extraction were discussed through single factor test and orthogonal test.The best extraction condition was that extracting time 2h,material liquid ratio 1:25(g/mL),extracted 3 times and extraction temperature of 90?.The extraction rate of chrysanthemum polysaccharides was 20.82 ±0.22%under the best condition.The content and molecular weight of chrysanthemum polysaccharide were determined.The results showed that the content was 91.11 ±0.37%,and the molecular weight was 2.98 × 103kDa.The chrysanthemum polysaccharide color reaction and specific rotation determination results show that the chrysanthemum polysaccharide components are insoluble in acetone,ethanol,ethyl acetate,n-butyl alcohol and other organic solvents,It is not including reducing sugar,polyphenols and starchy material.The specific rotation is +155 °.The chromatography and monosaccharide composition analysis results show that the chrysanthemum polysaccharide composed of rhamnose,arabinose,glucose,mannose,galacturonic acid and glucuronic acid,the mole ratio of 1:070:1.14:1.48:0.81:1.67;The structure of chrysanthemum polysaccharide was characterized by NMR,FT-IR analysis,periodate oxidation,Smith degradation analysis.The results indicated that the chrysanthemum polysaccharide is alpha anomeric configuration.Finally the activity of Imperial chrysanthemum polysaccharide was analyzed.The results showed that chrysanthemum polysaccharides can promote the acid production of lactic acid bacteria(Bulgarian bacillus)fermentation and promote the growth and proliferation of lactic acid bacteria effectively.Chrysanthemum polysaccharide yoghurt formula is optimized,the results show that the amount of white sugar was 8%,the added amount of polysaccharide was 0.5%,the inoculation amount was 3%,and the fermentation time was 5h.In this condition,the color of chrysanthemum polysaccharide yoghurt was milky white,the texture was even,the curd condition was good,the flavor was strong,the flavor was rich and smooth,and the flavor was characteristic of chrysanthemum.
