This topic mainly explores the changes of physical and chemical properties of the waste beer yeast extract under mannanase treatment,antioxidant capacity and yeast cell wall morphology were analyzed to make sure the best way of adding enzyme,then investigate the influence of intervention on STZ induced diabetic mice.Based on the beer yeast cell,this article studied the effect of beta-mannase on the the autolysis and product file of yeast cells.This study compared three ways of yeast autolysis.The first one is the autolysis of yeast without enzyme addition;the second one is the addition of enzyme during the autolysis;and the third one is addition of enzyme after the yeast cell autolysis.The results showed that,compared the first one,the method of enzyme addition promoted the autolysis of the yeast cells,and the autolysis led to the yeast cells becoming greatly shrunk with increased solid content in the liquid.Autolysis increased DPPH from 29.9%for without enzyme to 75.0%for sample with enzyme during autolysis and 77.8%for sample with enzyme after autolysis.Correspondingly,ABTS+ increased from 38.7%to 67.8%and 89.5%respectively.The molecular size was bigger in sample following autolysis without enzyme,The addition of enzyme reduced the molecular size.The data further confirmed that the use of enzyme increased solid content antioxidant capacity and reduced the molecular size of the substrates in the supernatant.The addition of enzyme following autolysis produced a narrowed molecular profile in the product,indicating a better method for utilizing the resource of yeast waste.This experiment regard kunming mice as the research object,aiming to study the influence of supernatant and precipitation which are from waste beer yeast cell(enzymatic treatment after the yeast cell autolysis)on glucolipid metabolism,oxidative stress and liver enzymes of diabetes mice induced by STZ.And using Quantitative Real-time PCR technology determine the effect of Intervention content on the mRNA transcription level of genes referred lipid metabolism,oxidative stress reaction in diabetic mice.This study also analyzed the differences of effect of supernatant and precipitation on related metabolic regulation of diabetic mice and concerned regulatory mechanism.Results of serum showed that physiological and biochemical indicators of mice serum,which suggest that the supernatant and precipitation could improve damage of metabolic disorders of diabetes mice to a certain extent,such as blood lipid metabolic disorders,oxidative stress injury and liver enzymes injury.It is notable that the supernatant intervention performed outstanding in regulating hyperglycemia and oxidative stress damage of diabetic mice,and it can significantly enhance the antioxidant capacity of the mice,balance diet and improve the health condition of mice.Result of Molecular biology experiment showed that determining the mRNA transcription level of related genes of lipid metabolism(PPARa,CPTla,HMG-COA,LDLR)and oxidative stress(Trxl,HIF-la,HO-1),research found that intervention can significantly improve the transcription level of lipid metabolism genes and oxidative stress genes in mice.This study also deeply analyzed the regulation pathways and mechanisms of intervention on related genes.All of these not only provides a strong evidence for the comprehensive utilization of beer waste yeast generated from the process of producing beer in the future,but also supply a new possibility for dietary intervention to alleviate the symptoms of diabetes.
