Objective To detect intracellular labile iron,novel fluorescent probes are designed and synthetized..Method(1)The fluorescent moiety of the original coumarin-based 3-hydroxypridin-4-one probes are replaced by dansyl to improve the hydrophilicity.(2)The original 3-hydroxypridin-4-ones probes are modified by adding an ester which can be hydrolyzed by the esterase in the cell,and the released carboxyl group can prevent the probe penetrating from the cell.Results With an increase of Fe(III)concentration,the fluorescence of the probe gradually decreased,the other metal ions have no significant influence to the probes.Conclusion The probe exhibits high sensitivity and selectivity towards Fe3+.They have moderate partition coefficient and readily penetrate cell membranes to detect intracellular iron pool.
