Chlorogenic acid was extracted from Eucommia ulmoides leaves mainly by water extraction,ethanol reflux,water extraction and ethanol precipitation.In this paper,chlorogenic acid extracted from Eucommia ulmoides leaves was optimize d by enzyme method and its technological parameters were optimized.Based on the extraction rate of chlorogenic acid as the evaluation criterion,three enzymes were selected to screen the enzymes for extracting chlorogenic acid from Eucom mia ulmoides Oliv.Leaves.After determining the enzymes,five factors,such as the ratio of material to liquid,the pH value of enzymatic hydrolysis,the amount of enzymatic enzyme,the time of enzymatic hydrolysis and the temperature of enzymatic hydrolysis,were selected for the single factor experiment.On the basi s of this,the optimum technological parameters of enzymatic extraction of chloro genic acid from Eucommia ulmoides Oliv.were determined by response surface analysis with appropriate factors and levels.The results showed that the extractio n rate of chlorogenic acid by cellulase was higher than that by other two enzym es.Response surface analysis showed that when the pH value was 4.5,the amou nt of cellulase was 553 U/g,the extraction temperature was 50 C,the ratio of material to liquid was 1:8,the extraction time was 75 min,and the extraction ti me was 75 min.The extraction rate of chlorogenic acid was the highest,reachin g 53.16mg/g.In addition,the antioxidant activity of eucommia chlorogenic acid was studie d by measuring the extraction of eucommia chlorogenic acid of different purity,the standard product of chlorogenic acid and the removal ability of Vc to DPPH free radical,reduction ability and superoxide anion?O2-·?.At the same time,fa ctors such as temperature,pH,light,and common metal ions were used to deter mine the antioxidant activity of chlorogenic acid at known concentrations to obta in how to avoid adverse factors in practical production practices.The results sho w that Eucommia extract has strong antioxidant activity to DPPH and superoxide anionic radicals and has a good effective relationship with its concentration.Te mperature and common metal ion impurities do not have a great influence on th e antioxidant activity of chlorogenic acid.However,strong light and strong acid alkalinity have a great influence on the antioxidant activity of chlorogenic acid.Therefore,in the process of extraction and processing of chlorogenic acid,it sho uld be avoided as much as possible in the environment of strong light and stron g acid alkalinity.To ensure that chlorogenic acid can maintain high activity.
