Based on the study status of combining onion with grape,the main purpose of this thesis is to develop the healthy and functional food-wool onion compound microcapsules with the main functional components of the onion oil and the grape juice,and to study its lipid-lowering effect.This experiment has made breakthrough progress in the extraction technology and its use value of scallion oil,and has certain positive significance for the research and development of functional food industry.The test results are as follows:(1)The optimum process of enzymatic assisted organic solvent extraction of onion oil:In this experiment,the extraction process of Scallion Oil was studied.Through the comparison of test methods,it was determined that the enzymatically-assisted organic solvent extraction method was used to test the hairy onion oil extraction on the pretreatment stage.When the ratio of solid to liquid was 1:4,comparative experiments were conducted on the extraction of organic solvents such as absolute ethanol,petroleum ether,methylene chloride,and trichloromethane.The optimum extraction solvent determined based on the yield of the onion oil was chloroform.We selected the enzyme addition,the temperature of enzymatic hydrolysis and the time of hydrolysis to perform a single-factor test to determine the optimum range of process parameters.An orthogonal test was conducted on the conditions of enzymatic hydrolysis to obtain the optimal conditions for enzymatic hydrolysis: the enzyme addition is 0.03%.The enzymolysis temperature was 45°C,the enzymolysis time was 2 h,and the yield of green onion oil was 0.42%.The stable,pale yellow viscous liquid was obtained.(2)Monoglyceride concentration:In this study,the compounding ratio of scallion oil and grape juice was studied.Based on the anthocyanin intake of the green onion extract and grape,the approximate compounding ratio was calculated.The single factor test method was used to test the concentration of monoglyceride.According to the emulsion stability,the concentration of monoglyceride was 1.5%.(3)Optimum spray drying conditions for the development of composite microcapsules by optimized spray-drying of response surface:In the experimental process,Optimization of spray drying with response surface was used to develop the composite microcapsules.Using sodium alginate as the wall material and monoglyceride as the emulsifier,the effects of various parameters of spray drying on the microencapsulation efficiency were studied.We selected the inlet temperature,concentration of sodium alginate,and rotation speed to conduct a single-factor test.The best parameter range was determined and the response surface was optimized to obtain the best process: the inlet temperature is 140°C,the sodium alginate concentration is 1.6 %,and the speed is 19 r/min.According to the quality index of microcapsules,the moisture content is determined as 5%,the dissolution time is 36 s,the bulk density is 0.25 g/L,the fluidic angle of repose is 30°,and the particle size is 4-90?m,there is no pathogenic bacteria,and the composite microcapsules were uniform and stable with good fluidity.(4)In the study of lipid-lowering effects,60 SPF Kunming mice were used to establish a high-fat diet by feeding high-fat diet.After one week of feeding,the body weight of the model group and the blank group increased significantly,and the blood lipid levels increased greatly.The model was successfully established.The experimental results show that the effect of the compound microcapsule medium dose group on the mice's hypolipidemic effect is significantly higher than that of the model group.The organ index shows that the microcapsule has an antagonistic effect on the action of the mouse organ and hyperlipidemia,and can effectively relieve the organ damage from Hyperlipidemia,the minimum effective dose is 110 mg /d,the minimum intake is 1 / day in the No.4 capsule.According to the results of the rules and animal experiments in the "Functional Evaluation Procedure and Test Method for Health Food Functional Evaluation",the formula design of the compound microcapsules of Mao Rong Cong and grape juice is reasonable and the preparation properties are stable.The compound microcapsule pair High-fat model mice certain lipid-lowering effects,and have certain health care functions,with good market development prospects.
