In this study,the ovum of Rana dybowskii was taken as the research object,its fatty acid composition analysis and separation and purification,oxidation stability,pharmacological effects were studied.And it was further applied to the development of cosmetic,the quality evaluation of cosmetics was studied。The main contents of this thesis are as follows:1.The components of fatty acids,polyunsaturated fatty acid enrichment process and oxidation stability in the ovum oil of Rana dybowskii:Fatty acids in the ovum oil were prepared.Then,fatty acids were detected by GC-MS.Polyunsaturated fatty acids were separated and purified by urea package,and orthogonal test were done.The ovum oil was stored in different conditions for 0,6,12,18,24 and 30 days.In the end,the oxidation stability was determined.Results showed that 18 kinds of fatty acids were separated from the ovum oil and the total content of unsaturated fatty acids was 76.76%,and polyunsaturated fatty acids were 40.25%.The optimal conditions of polyunsaturated fatty acid enrichment process were urea-fat ratio of 2,inclusion temperature of-20℃and inclusion time of 15 h.The smallest change was foundwith peroxide value of ovum oil under the condition of-20℃and avoided light preservation.2.The antioxidant and moisturizing ability of fatty acids in ovum oil of Rana dybowskii:Fatty acids in ovum oil of Rana dybowskii was regarded as the object of the study.The scavenging effects of DPPH·free radicals,ABTS·free radicals and hydroxyl radicals were determined respectively,and its reduction was also determined.Its water retention performance in vitro was measured.In the antioxidant experiments,the IC50 of fatty acids in ovum oil of Rana dybowskii in DPPH·free radical scavenging was 5.08 mg/mL and the maximum clearance rate was 80%.The IC50 of ABTS+·free radical scavenging was 9.84 mg/mL and the maximum clearance was 71.43%.The IC50 for scavenging hydroxyl radicals was 7.87 mg/mL.And its reducing power also increased with the increase of the solution concentration of fatty acids in ovum oil of Rana dybowskii.In the water retention experiment in vitro,the moisturizing ability of fatty acids in ovum oil of Rana dybowskii were significantly lower than that of hyaluronic acid,glycerol and 1,2-propanediol between 0 and 12 h,Its moisturizing ability had not significant difference with glycerol and 1,2-propylene glycol at 24h.The results showed that the fatty acids in ovum oil of Rana dybowskii had lasting moisturizing effect.According to these two properties,it can be applied to antioxidants and moisturizing cosmetics further.3.Development of cosmetics of ovum of Rana dybowskii:The cosmetics formulation and preparation process were screened,and the cosmetics matrix was scored according to the comprehensive performance score method table.The formula and preparation process of the lotion was established with the score as the index.The formulation of skin care milk was finally determined to be 2%of fatty acid,3%of glycerol monostearate,1.5%of cetyl alcohol,1%of octadecanol,4%of glycerol,3%of petrolatum,3%of Tween-80,5%of triethanolamine,0.75%of totaxin.The best preparation conditions for the emulsification temperature 70℃,emulsification time 40min,stirring speed 800r/min.4.The quality standards of the cosmetic of ovum from Rana dybowskii:Sensory index(appearance and aroma),physical and chemical indexes(heat resistance,cold resistance,pH value),hygienic index(mercury,arsenic,Determination of Lead,Total Bacteria,Detection of Staphylococcus Escherichia,Pseudomonas aeruginosa,Staphylococcus aureus),safety assessment(acute and repeated skin irritation test)and accelerated test were conducted.After testing,the indicators are in the standard value range;mice did not appear edema and erythema in the Acute and repeated skin irritation test.After accelerating experiment,there were no basically changes in the cosmetic.Based on the above research and analysis,ovum from Rana dybowskii contain a large amount of polyunsaturated fatty acid components,which have antioxidant and moisturizing properties,it’s suitable for the development of cosmetics.It also has good skin care effects.And the cosmetic is in line with cosmetics hygiene,microbiology and safety evaluation standards.
