Phytosterol is a kind of very important steroidal compound,which is 4-No methyl sterol containing ?-sitosterol,Campesterol,Stigmasterol and Brassicasterol etc.Most of them come from deodorization material which can inhibit the absorption of cholesterol for body,and also it is an active ingredient for reducing the functionality of serum cholesterol.However,the solubility of free phytosterol is low in water and greases;these characters limited the scope of application of phytosterol seriously.Numbers of experimental studies have shown that phytosterol esters have better ester solubility and higher cholesterol-lowering effect than free phytosterol,and it also can be used as functional food additives to added into food in order to achieve better application effect.This thesis do a systematically research in preparation?extraction techniques and analytical methods of phytosterol esters for broadening the application area of phytosterol.Compared with the previous results of research,there are some advantages of this method in this thesis,for example,the preparation is easier,solvent extraction is easier to restore and lower toxicity,which reduces the pollution of environment and created a new analytical method with high product content.The major findings of this thesis are as follows:1.The preparation technology of phytosterol estersWe compound phytosterol tea oil esters by the methods of ester transesterification of phytosterol and tea seed oil methyl ester,and we choose the fatty acid of tea seed oil.The tea seed oil has a good health care function which can improve the efficacy of the products.Sodium hydroxide compound the tea seed oil methyl ester as the catalyst which amount 5%of methyl ester,and then beat them up at a temperature of 68? with water by machine,and the reaction time is 4 hours.We evaporated them by rotating vacuum and got the tea seed oil methyl ester by delamination.We used the transesterification method of phytosterol and tea seed oil methyl ester to synthesize the phytosterol esters,and then we screened the transesterification reaction out by the method of orthogonal test.The best technology condition was n(tea seed oil methyl ester):n(phytosterol)=2:1,the quantity of catalyst addition was 3%of the quality of phytosterol,and the reaction time was 4 hours,the ester rate could reach 63.4%.2.The extraction of phytosterol estersThe experiment compared by many extraction processes,such as recrystallization and extraction,and the best technology was the extraction process.We could get the phytosterol esters with 93.3%yield by the process after twice washed,leached by methanol and n-hexane and refined processes.This extraction process was simple and the solvent was easy to reclaim.The extraction method with methanol and n-hexane was economical and novel.
