ObjectiveTo prepare terbinafine hydrochloride transfersomes with high encapsulation efficiency,appropriate drug loading and good stability that will meet the requirements of transfersomes.To optimize transfersomes with high permeation rate by the transdermal test in vitro.And then check the in vivo pharmacokinetic of the transfersomes gel prepared.MethodsUse the HPLC to establish the method of the determination of content and encapsulation efficiency of terbinafine hydrochloride transfersomes,then,study on the nature of the drug terbinafine hydrochloride before prescriptions.Take encapsulation efficiency as the main indicator,investgate the formulations and the processes of preparation.Use the same processes to prepare transfersomes with different surfactants,and optimize the best formulation with the transdermal test in vitro.Then prepare it as a gel and give a brief evaluation to it.At last,use the microdialysis technology to study the pharmacokinetic properties through the skin of this formulation.ResultsMean diameter of the terbinafine hydrochloride transfersomes was 69.6nm and the encapsulation efficiency was 95.4%,the content of the gel is 4.45mg/g,The accumulated permeation volume in 12h of the transfersomes gel was 88.52± 4.06ug · cm2,and the intracutaneous drug detention w as 94.38 ± 5.26 ug · cm-2,the results of pharmacokinetic studies showed the Cmax?AUC were apparently higher than the cream on sale.ConclusionThe terbinafine hydrochloride transfersomes which we prepared can be highly absorbed by the skin,the absorption rate is significantly more than the commercial creams either in the transdermal test in vitro or in the pharmacokinetic studies.The terbinafine hydrochloride transfersomes is a newly found efficient formulation for local external use.It has abslute advantages being compaired with the traditional prescriptions.So it has a very good application prospect.
