To explore the molecular mechanism of aroma's formation of flue-cured tobacco in our country,the main aroma matter composition and the relative metabolic pathways of flue-cured tobacco were comparatively analysised of four tipical provinces related to flue-cured such as Fujian,Yunnan,Henan and Guizhou.And using comparative proteomics,the study tried to expound the difference of aroma constituents in flue-cured tobacco and from the angle of the metabolic pathway to disscuss the molecular mechanism which leads to different aroma of flue-cured tobacco.The main results are as follows:1.The analysis of main chemical component in 12 samples of tobacco leave from four different typicality ecological regions shows that most of chemical component in the samples are within the scope of tobacco's quality indexes.But there are large difference from different typicality ecological regions,including both different and same aroma from different producing areas.All the chemical composition of tobacco leaves from Fujian and Yunan provinces reached extremely significant level.2.The aroma constituents from four typical areas show their significant difference.The contents of cembrane degradation and neophytadiene of flue-cured tobacco leaf in different typical ecological areas also show their significant difference,and Henan provinec has the highest content.All of these shows that the ecological environment had a great impact on the aroma constituents flue tobacco produced.3.The six groups of comparative protein profiles were established according to the tobacco leaves from 4 typical ecological areas,then 72 protein spots in different expression were found and 67 proteins in high matches were identified of those including tobacco carbon metabolism,post,plastid pigment degradation metabolic pathway,protein synthesis,and many other aspects.4.This research has a tentative explore to the difference of metabolic pathways of flue-cured tobacco in different typicality ecological regions and the relations among the different chemical conponents.The result of the study was that five raise proteins associated with plastid pigment degradation pathways in Fujian province were identified.They were Fructose-bisphosphate aldolase(S44),Ascorbate peroxidase(48),Triosephosphate isomerase,cytosolic(61)Hydroxy-pyruvate reductase(62),Pyruvate orthophosphate dikinase(66),the five rise proteins in Fujian which are benefitial to the formations of the aroma of plastid pigment degradation substance.9 raise proteins associated with Aldehyde ketone secondary metabolism related matter pathways were identified in Yunnan province,which were Quinone oxidoreductase protein(S28),Hydroxypyruvate reductase(S 19,S21),Fructose-bisphosphate aldolase(29),Phosphoglycerate kinase(31),Triosephosphate isomerase,chloroplastic(S38),Glyceraldehyde-3-phosphate dehydrogenase-A(39),Glyceraldehyde-3-phosphate dehydrogenase(S40,S41),NAD-malate dehydrogenase(S42),Fructose-bisphosphate aldolase(S43).These 9 rise proteins in Yunnan had Significant impact on the forming of the tobacco aldehydes,and ketones substance.Another five proteins associated with carbon metabolism and energy metabolismwere identified as well.Three raise proteins associated with Post type of metabolism pathways in Henan province were also identified which are Fructose-1,6-bisphosphatase,cytosolic(S32),Malate dehydrogenase,mitochondrial(34),Glyceraldehyde-3-phosphate dehydrogenase(S35),most content of those embrane degradatio are Terpenoid substances and shows cembrane degradatio had larger contribution to the flue-cured tobacco aroma formation in Henan province.Only four raise proteins in Guizhou province were identified,which were associated with energy metabolism and protein synthesis,not related to aroma substance metabolism.
