This article tried to prepare semi-interpenetrating polymer network hydrogels with KGM and Pullulan as the main raw material,in order to obtain intelligent hydrogels with excellent gel performance and a certain environmental sensitivity.Firstly,this article studied rheological properties of KGM and Pullulan system to preliminarily determine the better blending conditions;by using semi-interpenetrating polymer network(semi-IPN)technology,prepared KGM and Pullulan semi-interpenetrating polymer network hydrogels,and designed response surface methodology optimization with gel properties including gel strength,springiness,cohesiveness,chewiness,resilience,sensory evaluation and comprehensive evaluation as the response values;studied its environmental sensitivity(pH value,temperature,ionic strength),and laid the theoretical and experimental basis for further research of application as drug release carrier.The main results were as follows.1.Study of rheological properties of KGM and Pullulan systemThe addition of Konjac Gucomannan could significantly increase the viscosity and dynamic modulus of the system.The viscosity and dynamic modulus increased with appropriate amount of pullulan adding,whose concentration was less than 2.0%,and decreased significantly when above 2.0%.The addition of pullulan can improve the stability and the damping performance of the system.In alkaline conditions,medium viscosity,good dispersibility and stability,better gelation resistance and excellent damping performance could be obtained within the scope of testing,when KGM concentration was 1.0%?1.5%and PULL adding was 2.0%?4.0%in the system.2.The response surface methodology optimization of KGM and Pullulan semi-interpenetrating polymer network hydrogelsWhen concentration of KGM was 1.515%,concentration of PULL was 3.187%,concentration of Na2CO3 was 0.914%,hydrogels could get comprehensive evaluation value,that is gel strength was 127.394 g,springiness was 127.394,cohesiveness was 544.394 g,chewiness was 0.519,resilience was 0.223,sensory value was 9.615.3.The study of environment sensitivity of KGM and Pullulan semi-interpenetrating polymer network hydrogelsAs the temperature increases,the equilibrium swelling ratio of hydrogels showed a trend of increase first and then declined significantly with obvious temperature sensitivity;with the increase of pH,the swelling ratio of hydrogels showed a trend of obvious decline after rising first with apparent pH value sensitivity;the equilibrium swelling ratio decreased with the increase of ionic strength,showing nice ionic strength sensitivity;showed excellent temperature and pH impulse response,laid the tiheoretical and experimental basis for further research of application as drug release carrier.4.The structure characterization of KGM and Pullulan semi-interpenetrating polymer network hydrogelsa.FT-IR analysisCompared with KGM gel,O-H stretching vibration absorption peaks of KGM and Pullulan semi-interpenetrating polymer network hydrogels move to low band,which means bonding force of KGM and Pullulan semi-interpenetrating polymer network hydrogels hydrogen was strengthened,so KGM and Pullulan have the blending compatibility.b.The microstructure observationThe texture of KGM gel was relatively loose,shallow and closed the holes inside,and the hole is bigger,the width is thinner.What's more,the surface is covered with wrinkles.With the increase of concentration of PULL,the structure of semi-interpenetrating polymer network hydrogels was more compact,the surface of micro hole was smoother,narrower aperture,quantity increased,and specific surface area increased obviously,and formed clear water channels.
