In this paper,identify the strain with the ability to product flocculant D6,through 16SrDNA and physiological and biochemical tests to detect it to species.The flocculating activity of the culture fluid were measured to determine the distribution.Determine the main component of flocculant with color reaction,infrared scanning and UV scan.Purified the microbial flocculants and admeasure dry weight.To determine the optimal condition of flocculation by orthogonal test and determination of the optimum flocculation conditions on the removal of various pollutants.Study optimum parameters for slaughter wastewater treatment in the coagulation equipment on the foundation of best flocculation condition and put forward several suggestions according to the test results.After the test procedure,the final conclusion is as follows:(1)According to physiological and biochemical tests of microbial flocculant producing strain D6,combined with 16SrDNA sequencing results,determine the microbial flocculant producing strain D6 as Bacillus muralis.(2)Test results of the flocculating activity distribution showed that the fermentation liquid supernatant after centrifugation had the highest flocculating activity,reached 98.2%,the lowest flocculating activity for the bacterial suspension,only 66.8%.It proved that the activity components of flocculating was microbial metabolism of extracellular products instead of thallus itself.(3)Biuret reagent has no color change,Molish,anthrone and Benidict reagent were color change in the color reaction,and according to the infrared scanning and UV scanning results show that the main components of the microbial flocculant contained carbohydrate and non protein substances.The microbial flocculant was purified by acetone method,a rate of 11.06%,the total sugar content was 40.77mg/mL,accounted for 85.71%of the dry weight.(4)The microbial flocculant had good thermal stability,pH stability.Continued heating at 100? 35min,the microbial flocculant flocculation rate almost no decline,and in alkaline conditions,flocculation rate of flocculation was better than acidic conditions.(5)The optimum flocculation conditions of the microbial flocculant in slaughter wastewater was that microbial flocculant dosage was 25 mL/L,and the dosage of 1%CaC12 was 55 mL/L,pH 8.The slaughterhouse wastewater flocculation rate was 95.69%under the condition.(6)The optimum flocculation parameter during the purification process of slaughterhouse wastewater in the coagulation equipment was that the inlet velocity was 40L/h,and the microbial flocculant dosing pump was at the speed of 25?35r/min,1%CaCl2 dosing pump was at the speed of 50?60r/min.
