Mallotus barbatus,a member of Mallotus Lour.,belonging to Euphorbiacea,mainly distributes in Guangxi,Hubei,Hunan,Guangdong,Sichuan,Guizhou,Yunnan and other provinces of China.The roots of this plant be used as treatment agents of lung heat spit blood,damp-heat diarrhea,urine tube pain,indigestion and urethritis,and so on.Currently,few researches about Mallotus barbatus have been reported either at home or abroad.With the purpose of having a comprehensive understanding on the chemical composition of this plant,the constituents and anti-inflammatory activity of ethyl acetate part of 75%EtOH extract from the stem of Mallotus barbatus have been researched.In the dissertation,the ethyl acetate fractions of 75%ethanol extract from the stem of Mallotus barbatus was isolated using silica gel column chromatography(CC),MCI CC,Sephadex LH-20 CC,recrystal lizati on,silica gel preparative TLC CC,semi-preparative CC and preparative high performance liquid chromatograph(HPLC)CC.30 compounds were isolated,and 26 of them were identified by spectrometric data of NMR,U V,and MS.They are 1,1'-oxybis(2,4-ditert-butylbenzene)(1),2,2'-oxybis(1,4-ditert-butylbenzene)(2),taraxerone(3),?-sitosterol(4),?-homo-amyrin acetate(5),iup-20(29)-en-3b,30-diol(6),30-Hydroxylup-20(29)-en-3-on(7),acetyl aleuritolic acid(8).blumenol B(9),kaempferol(10),quercetin(11),kanzakiflavone-2(12),obovatin(13),3,5,7,4-tetramethoxyflavone(14),luteolin(15),scopoletin(16),isoscopoletin(17),3,4-Dihydroxybenzoic acid(18),terephthalic acid diethyl ester(19),trans-ferulic acid(20),vanillic acid(21),5-Methoxy-2-methyl-benzene-1,3-diol(22),galic acid(23),ethyl gallate(24),stearic acid(25).and ethyl stearate(26).According to the data-base of SciFinder Scholar,compounds 1,2,4,5,6,7,8,9,12,14,15,17,20,and 21 were firstly isolated from Mallotus barbatus.Compound 1 was firstly isolated from the natural products.The inhibition of NO production by LPS-induceds in RAW264.7 cells of six flavonaid compounds was evaluated.The results showed that compounds 10,11,12 and 14 could inhibit the NO production in RAW264.7 cells.The most active compound 10 exhibited best inhibition of NO production with IC50 values was 21.62 ?M.
