Because of the excellent health care function,Tremella aurantialba polysaccharide attracts more and more attention.But the extraction technology of Tremella aurantialba polysaccharide is not satisfactory.This research was carried out to optimize the protocol for extraction of polysaccharide from fruiting bodies of Tremella aurantialba.This research used single factor experiment and response surface method to optimize the technology of polysaccharides extraction from fruiting bodies of Tremella aurantialba.The single factors were water-feed ratio,extraction time,extraction temperature,grain size,extraction times.The response surface method is based on these optimized factors.In the single factor experiments,the water-feed ratio gradient were 100:1,200:1,300:1,400:1,500:1(mL:g);the extraction time gradient were 10,20,30,40,50 min;the extraction temperature gradient were 50,60,70,80,90,100?;the grain size gradient were 40,60,80,100,120,140 meshes;the extraction times gradient were 1,2,3 times.Polysaccharide extraction rate was figured out by phenol-sulfuric acid method.Every single factor experiment got an optimum range from each gradient,these ranges were used to have a response surface experiment.The single factor experiments indicated that the optimum water-feed ratio was 300:1(mL:g),the optimum extraction time was 50 min,the optimum extraction temperature was 90?,the optimum grain size was 120 meshes,the optimum extraction times was once.The result of response surface experiment predicted that the optimum conditions were water-feed ratio 347.38:1(mL:g),extraction time 52.71 min,extraction temperature 92.32 ?,grain size 116.79 meshes.Under these conditions the polysaccharide extraction rate was12.78%.Taking the realistic production condition into consideration,the verification test used water-feed ratio 350:1(mL:g),extraction time 53 min,extraction temperature 92?,grain size 120 meshes to test the reliability of the prediction.The polysaccharide extraction rate was 12.69%,0.09%lower than the theoretical value,so the optimal extraction process was reliable.
