Walnuts have high nutritional value.Walnut kernel pellicle contains most polyphenols of active ingredients.Therefore,the extraction of polyphenols from walnut kernel pellicle has great significance for the development of functional foods,the development of traditional Chinese medicine and enhancing the value-added products.Subject to the walnut in fen yang of Shanxi province as raw materials,analyse the composition and structure of the walnut kernel pellicle,and then research on the activity of immune,in vitro anti-tumor and in vivo anti-tumor.Main results are as follows:(1)Removal of Walnut seed coat using a new method of salt solution dipping method,measured the contents of water,ash,protein,fat,zinc and calcium of walnut kernel,The content of Zn in walnut seed coat was higher than that in the common walnut kernel,which was 5.78 times higher than that of walnut kernel.And Ca in was 2.29 times(2)The extraction rate of crude extract of walnut seed coat was 14.34%,and the content of polyphenol in walnut kernel was 19.10%.After purification by NKA-9 macroporous resin,the purity of the sample was about 65.2%.(3)UV-visible absorption spectra shows that there was a characteristic absorption peak at 268 nm and 360 nm in the extract from the seed coat,which indicated that polyphenols were extracted from walnut kernel.Infrared detection revealed that the walnut seed coat extract contained many characteristic functional groups of polyphenols,and the compounds were identified as polyphenols.(4)Research on immune activity of polyphenol in walnut kernels the results showed that walnut seed coat polyphenols immunocompromised mice induced a certain extent.The polyphenols of walnut seed coat could significantly improve the organ index of immunosuppressed mice,increase the carbon clearance index of immunosuppressed mice,increase the rate of lymphocyte proliferation,and maintain the blood balance of immunosuppressed mice to some extent Immunosuppressive mice immunoglobulin content,enhance the activity of LDH and ACP enzyme,thereby enhancing the immunosuppressed mice immune function.(5)In vitro antitumor activity of polyphenols from walnut kernel towards H22 hepatocellular carcinoma cells with different concentrations and treatment time was studied.The inhibition rate of 50?g/mL walnut seed coat polyphenol on H22 hepatocellular carcinoma cells was low,only about 15%There was no significant difference between 200?g/mL and 400?g/mL,the maximal inhibitory rate was reached at 72h.(6)Selecting the H22-bearing mice,and gavaging the mice,In vivo antitumor activity of polyphenols from walnut kernel was studied.The results showed that the polyphenols of walnut kernel could protect the immune organs.The results showed that the polyphenol of walnut seed coat could significantly enhance the proliferation activity of spleen T cells and B cells and increase the proportion of splenic T lymphocytes in spleen lymphocytes of each group.The results showed that the polyphenols of walnut seed coat could effectively maintain the balance of various cells in the blood of tumor-bearing mice and enhance the activity of T cells and B cells in vivo.By observing the H&E staining of solid tumors in mice,it was found that the polyphenols in the walnut kernel seed coat could make the morphological abnormality of the tumor cells appear in the solid part of the mice,and the necrosis phenomenon appeared in the tumor cells,such as nuclear pyknosis and nucleus disappearance.The anti-tumor immunity of mice was improved.
