The solubility of protein and the application value of protein can be enhanced by hydrolysis.However,the process of preparation and purification of natural enzyme with hydrolysis is complicated and the cost is high,so the simulation of protease has been widely concern.Cancer poses a great threat to the health of human beings,there are some disadvantages in the chemotherapy drugs,such as poor efficacy and poor selectivity.Therefore,the research of drugs with anticancer activity and selectivity has become a hotspot in the field of Medicine.The existing protease has the hydrolysis effect under the conditions of alkaline and high temperature,which limits its application in various fields,especially in the field of biomedicine.In this thesis,CQDs/Cu2O?carbon quantum dots/cuprous oxide?was used to simulate bovine serum albumin and casein protease enzyme under neutral conditions.Based on the mimetic enzyme activity of CQDs/Cu2O,further study on the application of CQDs/Cu2O material in anticancer.The effects of reaction temperature,reaction time,pH on hydrolysis of BSA and casien were investigated.CQDs/Cu2O has the catalytic activity of hydrolyzing BSA at 37? and the neutral condition of pH 7.4,and prolonging the reaction time or increasing the temperature can improve the hydrolysis rate.The first order reaction rate constant k0 is 3.5x10-4 S-1,Kcat is about 3.93×10-4 s1.Under the condition of 70? and pH 7.4,CQDs/Cu2O is also able to hydrolyze the casien with strong stability,and the first order reaction rate constant k0 is 7.8×10-4 s-1,Kcat is 10.78x10-4s-1.By electrophoresis experiment and the XRD spectrum of the materials before and after the reaction,the hydrolysis of BSA and casien can maintain good catalytic activity after ten times.The anticancer activity of CQDs/Cu2O was investigated by in vitro cell experiments.CQDs/Cu2O is able to inhibit the proliferation of cancer cells,the inhibition of cancer cells stronger than normal cells.The inhibitory effect of CQDs/Cu2O on human ovarian cancer?SKOV3?cells was the strongest.CQDs/Cu2O can induce the apoptosis of SKOV3 cells and block the cells in S phase,which can reduce the fluorescence intensity of F-actin cells.CQDs/Cu2O inhibits cell migration by controlling the expression of matrix metalloproteinases?MMP-2?protein in SKOV3 cells.CQDs/Cu2O was able to inhibit angiogenesis,the inhibition was higher than commercial cuprous oxide and tyrosine kinase?SU5416?.
