Protease is a type of catalytically active protein that is involved in all life activities and has a very important application in the fields of washing,medicine,food,diagnosis and treatment.Protease wide applications,however,the high cost of natural protease,the reaction conditions are harsh,poor stability,these disadvantages limit the application of the protease.In addition,the diagnosis and treatment of cancer is also a major problem in medical research.Ovarian cancer is one of the most common cancers among women.Therefore,with the continuous improvement of the level of science and technology and research,some new nanomaterials have been applied to the research of analog protease mimics and anti-cancer fieldsIn this paper,magnetic ferric acid copper(CuFe2O4)was used as a research object to explore the activity of the mimics protease and its effect on human ovarian cancer cells.The main contents are as follows:(1)CuFe2O4 material for the hydrolysis of bovine serum albumin(BSA)and casein to explore the effect of reaction time,temperature and pH on the effect of hydrolysis,the experimental results show that in pH hydrolysis under neutral conditions is the best with the increase of temperature and time,the effect of hydrolysis increased.The biggest advantage is that CuFe2O4 has good magnetic effect and stability,and is beneficial to the recovery and utilization of materials.(2)CuFe2O4 material for mouse embryonic fibroblasts(BABL-3T3),human ovarian cancer cells(SKOV3),vascular endothelial cells(HUVEC),colorectal cancer cells(HT-29)and human non-small cell lung cancer cells(A549)Inhibition of the proliferation of the test,calculated CuFe2O4 on different tumor cells,half inhibition(IC50).The results showed that CuFe2O4 was more toxic to the four tumor cells than normal cells,but CuFe2O4 had the strongest inhibitory effect on the proliferation of SKOV3.The IC500 value was 3.1μg mL-1 at 24 hours and did not change with the increase of the action time.(3)Select the best inhibitory effect of SKOV3 as the research object.By Migration,apoptosis,and Western Blot analysis,the results showed that CuFe2O4inhibited SKOV3 cell migration by down regulating the expression of matrix metalloproteinase MMP-9,MMP-2 and actin actin.AO/EB staining and its flow analysis further showed that CuFe2O4 could induce apoptosis in SKOV3.
