L27(313)full factorial design was used to construct microflora degrading phenol.The analysis of variance(ANOVA)results showed that sp.C1 was non-significant factor,but Bacillus sp.F6 and Arthrobacter sp.E2 were significant factor;two way interactions such as Bacillus sp.F6:Arthrobacter sp.E2 were the most significant interaction,but Klebsiella sp.C1:Bacillus sp.F6 or Klebsiella sp.C1:Arthrobacter sp.E2 were non-significant.The volume pronortion of optimal flora wasV(Klebsiella sp.C1):V(Bacillus sp.F6):V(Arthrobacter sp.E2)=2:1:3.The characteristics of mixed microflora JF degrading phenol and the effects of environmental factors on the ability of JF degrading phenol were performed in sealed condition.Under the influence of each growth factors,The phenol was degraded most efficiently by microflora in log phase.The optimized growth conditions of microflora JF and degrading phenol were:36?,pH 8.0,salt concentration 0.5 g·L-1.and the suitable conditions for the phenol efficiently degraded by microflora JF were:pH 4.0?10.0,temperature 15?45?,salinity?60 g·L-1 NaCl at any initial phenol concentrations ranged from 100 to 2500 mg·L-1.The result indicated that Microflora JF could tolerate the concentration of phenol up to 2500 mg·L-1.Microflora JF could tolerate KCl,K3PO4,NaNO3,Na2SO4,Na2CO3,CH3COONa which exist in usual wasterwater.The flora could grow with utilizing carbon sources lactose,glucose,sucrose,maltose,fructose,molasses,etc.and nitrogen sources casein,trypsin,yeast extract,ammonium chloride,ammonium nitrate,ammonium sulphate,ammonium acetate,ammonium phosphate,etc.But the growth of flora was inhibited by carbon source oat bran or nitrogen sources ammonium citrate,ammonium molybdate.Microflora JF,which is able to biodegrade mang kinds of aromatic compounds,could utilize phenolphthalein,chlorobenene,toluene,styrene,catechol,resorcin,hydroquinone,pyrogallol,hydroxyquinol,phloroglucinol etc.as its sole carbon source to support metabolism and grow proliferation,but couldn't use naphthalene or phenylacetaldehyde.In contrast to Klebsiella sp.Cl,Bacillus sp.F6,Arthrobacter sp.E2,microflora JF has more advantages in tolerating acid-base properties,high salt,limiting temperature of microorganism culture,high phenol concentration of phenolic wastewater.Microflora JF could be more widely utilize various of salts,carbon sources,nitrogen source etc.as energy to grow and phenol degradation.The flora could utilize more various of aromatic compounds as its sole carbon source to grow.Therefore,in contrast to pure strain(Klebsiella sp.Cl,Bacillus sp.F6,Arthrobacter sp.E2),microflora JF had more advantages in bioremediation of phenol contaminated environment.The effects of extreme environmental factors on phenol degradation were investigated.Microflora JF could withstand extreme ecological factors,such as phenol concentration 2200 mg·L-1,salt concentration 60 g·L-1,pH 5.0 and pH 9.0,no matter what the culture temperature(the optimal growth temperature or room temperature).The flora could keep efficient degradation of phenol at low temperature(15?)and changing of culture temperatures(15??25 ??35??25??15??25??35?...).Therefore,microflora JF may find great application in treatment of waterwast containing phenol.
