purpose:To investigate the stability of the non-steroidal anti-inflammatory drug indomethacin and its release in the human body.Method:The main drug for the indomethacin,accessories for the PVPP,PVP,MCC,SDS.The preparation of the pellets was carried out according to different proportions.The pellets were prepared by extrusion-roll method.The dissolution medium was set as HCl solution with pH=1.1 and PBS solution with pH=6.7 and pH=7.6 respectively.Respectively,to simulate the human body stomach,small intestine,colon environment.By changing the pH of the dissolution medium to simulate the environment in the human digestive tract,calculate the cumulative release of drugs in different environments to determine the main role of drugs.Result The main drug content was 40%,PVPP was 15%,PVP was 15%,and SDS was 0.5%.The indomethacin pellets prepared according to this formulation had the best powder properties and had the highest cumulative release at pH=7.6 PBS buffer solution.Conclusion The results show that extrusion speed,spheronization velocity and spheronization time are the key factors of the process.The preparation process and prescription composition are determined.The results show that the extrusion speed,roll speed and spheronization time are the key factors in the process.In the process of process factors,the extrusion rate has a great influence on the powder properties of the pellets.Indomethacin in the higher alkaline conditions,the higher the release rate,after several pre-experiments found that indomethacin pellets in the pH=6.8colony in the highest degree of release in the small intestine and stomach release degree decreased one by one,Indicating that indomethacin pellets are targeted and are a colonic localization agent.
