Shizhu county in Chongqing and Lichuan county in Hubei are the main producing areas of Rhizoma coptidis, accounting for80%of national output and called Coptis chinensis Franch. When harvesting Coptis chinensis Franch, Coptis chinensis Franch radix was produced, through collecting the adventitious roots, cleaning, and drying. Coptis chinensis Franch radix contains berberine, palmatine, coptisine, epiberberine, jatrorrhizine and other alkaloids, its medical efficacy was similar to Rhizoma coptidis, clearing heat and eliminating dampness, heat-clearing and detoxicating. Because the bitter taste, the oral usage of Coptis chinensis Franch radix is inconvenient. This experiment selects Coptis chinensis Franch radix produced in Shizhu, extracting its effective components, screening a suitable materials proportioning. Then Coptis chinensis Franch radix pellet cores were prepared by extrusion-spheronization and coated, and the coated pellet quality detective method was established.The extracting factors that affect the total alkaloids of Coptis chinensis Franch radix with berberine, palmatine, coptisine, epiberberine, jatrorrhizine, were analyzed. The results showed that the optimum extracting factors were:ethanol concentration70%, extraction time0.5h, extraction times3.The Coptis chinensis Franch radix pellets were prepared by extrusion spheronization. The proportion of starch to microcrystalline cellulose, the content of dextrin, the variety and content of adhesives, were assayed through single factor test for determination of pellet formula, with coefficient of recovery, bulk density, sphericity, friabibity, surface morphology and colligation score of pellets. The resulting formula was as follow:proportion of starch to microcrystalline cellulose7:3, the content of dextrin5%of total diluent,50ml of3%HPMC water solution, and loading dose as20%. The technologic factors that affect the formation of pellets including extrusion speed, spheronization speed and time were tested. The result showed the proper technologic parameters were:45rpm of extrusion speed,850rpm of rolling speed, and2min of rolling time.The multifunctional rolling machine was used to coat the Coptis chinensis Franch radix pellets. The determined technologic parameters were:100g of feeding mass,120rpm of turnplate speed,25Hz of blast speed,40℃of coating temperature,0.1Mpa spray pressure and3ml·min-1iquid suppling speed. The effect of proportion of acrylic resin Ⅱ and Ⅲ, content of plasticizer and antistatic agent SDS, and the coating pellet weight gain were investigated. The resulting formula was:2:1the proportion of acrylic resin Ⅱ and Ⅲ,20%the content of Plasticizer in total polymers,0.3%SDS content and10%increase in coating weight.The UV-spectrophotometric method was established to investigate the release of Coptis chinensis Franch radix enteric-coated pellets, and High-performance liquid chromatography was applied to determine of berberine content in Coptis chinensis Franch radix enteric-coated pellets. The stability test of Coptis chinensis Franch radix enteric-coated pellets showed the enteric-coated pellets are sensitive to high humidity, high temperature and light, so the pellets should be stored in dark and cool place.
