The optimization of experimental conditions of the extraction of several main fructo oligosaccharides and their biological activity in yacon,and provides the reliable^ scientific and accurate basis for the study of fructo oligosaccharides have been studied in this paper.So the research of fructo oligosaccharides in yacon provides effective,convenient,fast experimental method for the further study has been provided.In the study of the color-protecting,which discusses the blanching treatment,extraction time and antioxidant oxidation degree of impact on the yacon blended pulp.Blanching temperature was chosen as 20,50,and 80? for blanching 5 min,experiments show that the most of the loss of enzyme activity was maken on 80?,Extracting time was chosen as 0.5,1,2,2.5,3 h,the experiment showed that the best time for the extraction was 2 h.EDTA-Na2,ascorbic acid and citric acid have beeb Selected as the three antioxidant.Experiments were used three different concentrations of antioxidants was carried out on the yacon blended pulp processing,by ultraviolet spectrophotometry puree oxidized degree,determine the antioxidant effect.The results showed that the concentration of 1%EDTA-Na2 oxidation effect is best.In addition,the concentration of the mixed use of orthogonal experiment analysis,determine the influencing factors for primary and secondary VC>EDTA-Na2>citric acid,the optimal combination of 1%ascorbic acid + 1%citric acid + 0.25%EDTA-Na2.In the study on the extraction of yacon fructo oligosaccharides,which mainly for extraction conditions of the three main types of fructooligosaccharides in yacon(1-Kestose,Nystose and 1-?-D-fructofuranosylnystose).Extracting for fructo oligosaccharides by ultrasonic assisted extraction and microwave assisted extraction respectively.The results show that:The maximum extraction of 1-Kestose,Nystose and 1F-fructofuranosylnystose were 20.72%,9.97%and 10.83%respectively by ultrasonic assisted extraction,which was under the condition of the ratio of material to liquid of 1:50(g·mL-1),Ultrasonic Power of 80W,ultrasonic time of 35min,temperature of 70?,while the maximum extraction were 25.81%,10.80%and 10.85%by microwave assisted extraction respectively with condition of the ratio of material to liquid of 1:50(g·mL-1),Ultrasonic Power of 500W,ultrasonic time of 35min,temperature of 50?.And microwave assisted extraction is more efficient,convenient and simple than ultrasonic assisted extraction.In the study on the hypoglycemic effect mechanism of fructo oligosaccharides in yacon,exploring the hypoglycemic effect mechanism of fructo oligosaccharides in yacon to mice through comparing based on the comparison group,negative control group,positive control group and fructo oligosaccharides group.The results showed that:yacon high concentration dose group can obviously reduce the blood glucose of diabetic mice,in that,yacon oligosaccharides can play a goog role in lowering blood glucose.
