To investigate the anti-cancer and anti-inflammatory activities of volatile components in six types of tea: white tea,oolong tea,yellow tea,pu-erh tea,black tea,and green tea.Crude volatile components were extracted by simultaneous distillation-extracion(SDE)and further isolated using chemical method and thin layer chromatography method(TLC).Hepa1c1c7 and RAW264.7 cell models were used to evaluate the anti-cancer and anti-inflammatory properties of crude aromas and separative products.Gas chromatography-mass spectrometry(GC-MS)was applied to analyze volatile components.The anti-cancer and anti-inflammatory activities of the main monomers,such as ?-ionone,were evaluated by Hepa1c1c7,HCT116,HepG2,MDA-MB-231,and RAW264.7.The anti-cancer activities of interactions between monomers and oridonin with monomers were also studied using HCT116 cell models.The results were as follows: 1.Anti-cancer and anti-inflammatory activities of crude aromas of six types of tea(1)The crude aromas of six types of tea exhibited strong QR inducing activities and possessed strong NO inhibitory activities.Their CD values were 6.25-100 ?g/m L.Their concentrations to achive 50% inhibition of NO were 25-100 ?g/mL.The anticancer activities of white tea,oolong tea,green tea,and Pu-erh tea were stronger than that of black tea,and black tea is stronger than that of yellow tea.The anti-inflammatory activities of Pu-er tea,green tea and yellow tea were stronger than that of white tea,black tea,and oolong tea.(2)GC-MS analysis showed that the highest relative content of the volatile components was heterocyclic oxygen in Pu-er tea,and alcohols in the other five kinds of tea.The components of the volatile components of all kinds of tea with relative content of more than 5% were analysized.The main component in Pu-erh tea was 1,2,3-trimethoxybenzene.Linalool and linalool oxides,geraniol,phytol,and ?-terpineol were the main components in green tea.Linalool and linalool oxides,geraniol,and phenethyl alcohol were the main components in white tea.Linalool,geraniol,3,7-dimethyl-1,5,7-octatrien-3-ol,trans-nerolidol,and indole were tha main components in oolong tea.Linalool and linalool oxides,geraniol,phytol,and ?-terpineol were the main components in yellow tea.2.Anti-cancer and anti-inflammatory activities of volatile components isolated by chemical method and TLC method from three types of tea(1)There were 11 kinds of components seperated by chemical method and TLC method were idetified using the Hepa1c1c7 cell model: neutral component,F6,F6-2,and F6-3 in pu-erh tea;neutral component,F4,F5,F6,F7,and F6-2 in black tea.neutral component in green tea.The CD values were less than 50 ?g/m L.(2)There were three kinds of components seperated by chemical method and TLC method were idetified using the RAW264.7 cell model: neutral component in pu-erh tea,black tea,and green tea.The concentrations to achive 50% inhibition of NO were 25-100 ?g/mL.(3)GC-MS analysis showed that 1,2,3-trimethoxybenzene,1,2,4-trimethoxybenzene,1,2-dimethoxybenzene,isophytol,isooctanol,?-ionone,2-pentadecanone,tetrahydro-2,2,6-trimethyl-6-vinyl-3-pyranone,benzophenone,nonanal,carvacrol,heptadecane,and hentriacontane were the main monomers in Pu-erh tea.Linalool phytol,?-ionone,phenylacetaldehyde,tetracontane,eicosane,1-nonadecene,and methyl hexadecanoate were the main monomers in black tea.Linalool,geraniol,phytol,and ?-terpineol were the main monomers in green tea.3.Anti-cancer and anti-inflammatory activities of monomersAmong the seven monomers,?-ionone can induce QR multiplication.1,2,3-trimethoxybenzene,1,2,4-trimethoxybenzene,1,2-dimethoxybenzene,?-ionone,and phytol can inhibite the production of NO.?-ionone,phytol,?-terpineol,and phytol can inhibit the proliferation of HCT116,MDA-MB-231,and HepG2 cells.Results showed that 7 monomers were more sensitive to HCT116 cell.4.The anti-cancer activities of interactions between monomers and oridoninThe strongest combination of synergistic inhibition effect of HCT116 cell proliferation between monomers was 1,2,3-trimethoxybenzene 864.55 ?g/mL + 1,2-dimethoxybenzene 1209.74 ?g/mL,the proliferation inhibition rate was 78%,CI value was 0.05.The strongest combination of synergistic inhibition effect of HCT116 cell proliferation between oridonin with monomers was phytol 172.46 ?g/mL + oridonin 1.87 ?g/mL,the proliferation inhibition rate was 92%,CI value was 0.32.
