Tremella fuciformis Berk,also known as white fungus,mainly contains proteins,carbohydrates,vitamins,and various amino acids for medicinal and tonic foods.This experiment mainly used spores as the experimental material,using ethanol precipitation and deproteination with Sevage method,isolated and purified three kinds of uniform Tremella fuciformis polysaccharide which named TPF-1?TPF-2?TPF-3,with the method of water dialysis,DEAE-fast flow column and futher purified through Sephadex-G100 column chromatography fractionating,and then its physicochemical properties and antioxidant activity were measured.The main findings are as follows:(1)By ITS identification,two strains of relatively high producing strains Tyc63 and Tr01 were screened out from the six original strains of Tyc63,Y11,Y2,Tr01,Tr21 and Tr9901.The optimum fermentation period was determined to be 6 days.(2)Three purified polysaccharides TPF-1,TPF-2 and TPF-3 were obtained through the separation and purification of Tyc63 crude polysaccharide.TPF-1 was white floc,and TPF-2 and TPF-3 were loose white.All three are odorless,easy to absorb moisture,soluble in water,poorly soluble in methanol,ethanol,acetone and n-butanol and other organic solvents;its high concentration of aqueous solution was viscous.(3)The results of FT-IR spectroscopy indicated that there are characteristic absorption peaks of sugars in the infrared spectra of the samples TPF-1,TPF-2,TPF-3.These groups of peaks can be used to determine initially that the sample is a polysaccharide compound;and Only 2 peaks appear in the region of 1100 cm~-1 to 1010 cm~-1,which should be furanosides.Among them,TPF-1 and TPF-2 are barley-type polysaccharides,and TPF-3 is a?-type polysaccharide.(4)By GC-MS analysis,TPF-1 is mainly composed of fucose,xylose,mannose,glucuronic acid,and the molar ratio of several monosaccharides is 9.25:23.4:16.05:37.1;The proportion of TPF-2 and TPF-3 was mainly rhamnose,arabinose,mannose,glucuronic acid,and galactose,and their proportions were 1.78:9.55:12.63:128.59:13.91 and 3.8:30.78:52.78:12.73:14.6.It can be inferred that the main chain of TPF-2 may be an acidic heteropolysaccharide with a main chain of glucuronic acid.TPF-3 may be an acidic heteropolysaccharide with a mannose backbone.Congo red experiment shows that TPF-1and TPF-3 contain triple helix structure while TPF-2 without triple helix structure.(5)The weight average(Mw)of TPF-1,TPF-2,and TPF-3 were 5189 kDa,171.6 kDa,and 661 kDa respectively.(6)The antioxidant experiments in vitro showed that both crude polysaccharides and purified polysaccharides had certain antioxidant capacity,but the scavenging effects of several free radicals were lower than those of the same standard VC standards.When the solution concentration was 1 mg/mL,the reducing power of ABTs~+free radicals by crude polysaccharides was 27.08%,and TPF-1,TPF-2 and TPF-3 were 15.28%,44%and 41.9%,respectively.For the scavenging ability of hydroxyl radicals,the clearance rate of Tyc63crude polysaccharide reached 42.5%when the solution concentration was 1 mg/mL,while the purified polysaccharide had weaker scavenging ability for hydroxyl radicals.(7)We developed three kinds of cosmetic products using extracellular polysaccharides:extracellular polysaccharide toner,lotion,and moisturizing cream.The three cosmetics have extremely low cytotoxicity and they can protect HSF from UVB-induced damage.
