Seabuckthorn is a kind of common medicine and food homologous plant,which has more nutritional value and biological activity.As one of the main producing areas of Hippophae rhamnoides in China,Xinjiang is relatively rich in Hippophae rhamnoides resources.Therefore,how to make better use of and promote seabuckthorn industry is an important problem to be solved.In this paper,seabuckthorn fruit was used as the research material.Seabuckthorn crude polysaccharides were extracted in water and precipitated with ethanol,then obtained by ultrasonic assisted method.Three purified components of polysaccharides were obtained by using two kinds of chromatographic columns.The monosaccharide composition,infrared spectrum,scanning electron microscope and content determination results of each component were analyzed.The structure characterization and biological activity of purified components of seabuckthorn polysaccharide were preliminarily discussed,which provided a theoretical basis for the development and production of seabuckthorn.The main results are as follows:(1)The crude polysaccharide of Hippophae rhamnoides was separated and purified by two kinds of chromatographic columns,and three kinds of purified components were obtained.The infrared spectra showed that all the three polysaccharides contained characteristic peaks of carbohydrate compounds.It was inferred that SBP-1s and SBP-2s belonged to ?-configuration polysaccharides,and SBP-3s belonged to ?-configuration polysaccharides.Scanning electron microscope results showed that SBP-1s was loose flocculent structure,not fully aggregated.SBP-2s was irregular cone-shaped accumulation,SBP-3s was rough sheet structure,there are dense cavities with dense structure.The results showed that the content of total sugar and uronic acid in SBP-3s was the highest.(2)The antioxidant activity of SBP-3s was evaluated by chemical method and cell model method.The results showed that: in the range of 0.2?1.0 mg/m L,SBP-3s had a positive regulatory effect on DPPH and ABTS free radical scavenging ability.The scavenging effect and reducing ability were similar to those of the positive control group.In addition,the survival rate of oxidized LO2 cells treated with SBP-3s was 79.57%,that is,SBP-3s could protect the oxidative damage of LO2 cells.(3)The immunomodulatory and antitumor effects of SBP-3s were evaluated by cell culture in vitro.The results showed that with the increase of SBP-3s concentration,it can increase the phagocytic capacity of macrophage RAW264.7,increase NO and TNF-? and IL-1?.It has a certain immunoregulatory effect.SBP-3s can also inhibit the proliferation,apoptosis and metastasis of hepatoma cells,and reduce the expression of key proteins in p38 signaling pathway of Hep-G2 cells.
