Studies On The Anti-alcohol Activity Of Total Flavonoids Extract From Seven Plant Materials

Wine culture,enjoyed by many Chinese people,is also one of the important element in Chinese diet culture.However,excessive or long-term consumption of alcohol can cause numerous adverse effects on human organs like stomach,liver and kidney etc,which will ultimately not only lead to a decline in quality of life personally,but also bring about harm to both family and society.Therefore,search for different methods and anti-alcohol substances which can resolve and relieve the discomfort caused by drinking has become a major concern for human beings.In this study,preliminary exploration testing the hangover abilities was performed on activity intervention materials including kudzu root(Puerariae radix),fructus aurantii(Citrus aurantium),flos sophorae(Sophora japonica),vine tea(Ampelopsis grossedentata),elsholtzia(Elsholtzia ciliate),lotus leaf(Nelumbo nucifera)and honeysuckle(Lonicera japonica),in which Valle & Hoch experiment and Blair & Bodley experiment were conducted to test their activation effect on alcohol dehydrogenase in vitro.Meanwhile,based on the established drunken mice model,behavioral experiments were performed to explore the anti-alcohol effect of them.Several physiological and biochemical indexes were measured afterwards.The anti-alcohol effects of those 7 kinds of total flavones were analyesd and compared to that of the Neptunus(an antialcoholismic drug available on the market).All data were analyzed using statistical software SPSS 20.0.The main results of this study can be summed up as following:(1)Raw materials were extracted twice by 70% ethanol assisted by ultrasonic for30 mins each time.The total flavonoids content of flos sophorae,vine tea,lotus leaf,kudzu root,fructus aurantii,honeysuckle and elsholtzia were measured and listed as follows:87.65 mg /g DW,121.18 mg /g DW,111.83 mg /g DW,32.33 mg /g DW,30.58 mg /g DW,101.53 mg /g DW,44.40 mg /g DW.(2)Vitro experiments indicated that honeysuckle and elsholtzia did not possess anti-alcohol effect.Results from righting reflex test indicated that total flavones of flos sophorae,kudzu root and vine tea,as well as Neptunus could significantly prolong the incubation period(P<0.01)and reduce the drunken sleeping time(P<0.01)of the drunken mice.Total flavones of lotus leaf could also remarkably prolong the incubation period of drunken mice(P<0.05),yet the effect on reducing the drunken sleeping time of mice(P<0.01)were milder than those of the total flavones above.In climbing experiment,administration of the flavonoids of flos sophorae,kudzu root,vine tea and Neptunus significantly prolonged the climbing time of drunk mice(extended the duration of drunken mice stayed in the network)(P<0.01).However,total flavonoids of fructus aurantii did not exhibited certain anti-alcohol effect.(3)Comparing to the significant reducing ethanol concentration in drunken mice serum(P<0.01)performance that total flavonoids of flos sophorae,kudzu root and vine tea,as well as Neptunus exhibited,total flavonoids of lotus leafs reducing effect is milder(P<0.05).Flos sophorae,kudzu root,vine tea and Neptunus can significantly increase activity of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH)(P<0.01)in drunken mice,directly promoting the positive metabolism of ethanol.But,lotus leaf can only significantly increase activity of alcohol dehydrogenase(ADH)and aldehyde dehydrogenase(ALDH)(P<0.05).(4)Vine tea can significantly reduce the activity of glutamic pyruvic transaminase(ALT)(P<0.01),but,flos sophorae,kudzu root and Neptunus can only significantly reduce the activity of glutamic pyruvic transaminase(ALT)(P<0.05).Flos sophorae,kudzu root,vine tea and Neptunus can significantly reduce the activity of glutamic oxalacetic transaminase(AST)(P<0.01)in drunken mice serum,but,lotus leaf can only significantly reduce the activity of glutamic oxalacetic transaminase(AST)(P<0.05)in drunken mice serum.Flos sophorae,kudzu root,vine tea,lotus leaf and Neptunus can significantly reduce the triglyceride(TG)content(P<0.01).While total flavonoids of vine tea could significantly reduce liver coefficient of drunken mice(P<0.01),the flavonoids of flos sophorae,kudzu root,lotus leaf and Neptunus can only reduce liver coefficient of drunken mice(P<0.05)in a milder way.(5)Flos sophorae,kudzu root and vine tea can significantly increase the content of glutathione(GSH)and the activity of superoxide dismutase(SOD)in drunken mice(P<0.01),but,lotus leaf and Neptunus can only significantly increase the content of glutathione(GSH)and the activity of superoxide dismutase(SOD)in drunken mice(P<0.05).Flos sophorae,kudzu root,vine tea and Neptunus also can significantly decrease the content of malondialdehyde(MDA)in liver in drunken mice(P<0.05).These results showed that total flavonoids extracted from fructus aurantii,elsholtzia and honeysuckle had no anti-alcohol effect.Total flavonoids of Kudzu root,flos sophorae,vine tea and lotus leaf,as well as Neptunus possess certain positive anti-alcohol effect,which can be sorted accordingly as vine tea,flos sophorae,kudzu root,Neptunus and lotus leaf(from superior to inferior).It is worth emphasizing that the anti-alcohol effects that the total flavonoids from vine tea,flos sophorae and kudzu root performed were better than that of the antialcoholismic drug currently available on the market.In this case,this study may serve as theoretical basis for future research on the development of anti-alcohol products and bring about innovation to the anti-alcohol market.
