Chromatographic separation and solid phase extraction,as the two most commonly-used techniques in separation and purification filed,play important roles in separation science.Its separation medium is the cores of meeting different separation and purification requirements,the preparation of separation medium is important to develope the separation and purification technology.The separation media could be devided into two categories,silica-and polymer-based matrix.The polymer materials as separation medium has attracted much attention in separation science,since their advantages of good biocompatibility,low non-specific adsorption and high p H stability.However,polymer materials always tend to have uncontrollable morphology,single pore structure,low separation efficiency and poor mechanical stability,which limit their applications in separation science to some extent.In order to synthesize the polymer materials with high selectivity,abundant pores,high permeability and controllable morphology,the sacrificial support method affords us lesson in this paper to expand the applications of polymer materials.This paper focused on the topic of "the preparation and applications of polymer-based separation media”,used sacrificial support method synthetizing two kinds of polymer materials,and applied them to protein chromatographic separation and miniaturized solid phase extraction.The main contents include the following three aspects:1)The preparation of penetrable poly(MAA-co-EDMA)microspheres and its chromatographic application in protein separation.The penetrable poly(methacrylic acid-co-ethylene glycol dimethacrylate)(poly(MAA-co-EDMA))microspheres were successfully synthesized used penetrable silica as the sacrificial support.Owing to the microspheres possessed copious mesopores and throughpores,large surface area,adjustable hydrophobicity and the cation-exchange ability,they were applied as chromatographic stationary phase to separate proteins in RPC,HIC and IEC modes,respeectively.Moreover,thanks to their throughpores,fast separation at low column backpressure could be achieved in these three modes.2)The preparation of benzophenones molecularly imprinted polymer materials and its application in sample extraction.The molecularly imprinted polymer whichusd silica as support and 2,4-dihydroxybenzophenone as the template(DHBP-MIP)was synthesized via sacrificial support method.Since the good selectivity and adsorption capacity,the DHBP-MIP was used as the sorbent to a miniaturized sorbent phase-based extraction device in the form of syringe filter holder(?-SPE-SFH-MIP)device.Then the ?-SPE-SFH-MIP device was applied to extract several BPs from real samples.Under the optimal extraction conditions,the developed ?-SPE-SFH-MIP procedure showed satisfactory extraction efficiency for target BPs from human urine and swimming pool water samples,so that it was promising in sample pretreatment for complex samples.3)The introduction and summary of the application of sacrificial support method for providing a reference for the further study.
