In recent years,plant nematode disease has become a serious threat to forestry production in C hina.Since its first outbreak in 1982 in Nanjing,Bursaphelenchus xylophilus?pine wood nematode disease?has caused enormous losses in C hina over the twenty years.Besides,it has also damaged natural landscape and ecological environment,becoming a serious threat to rich pinewood forest in C hina.For now,though commercial nematocides has effectively prevented and treated nematode diseases,since it is of high toxicity and the nematode's strong drug resistance to it,to develop a new kind of nematocides has become an urgent issue.The rich secondary metabolites make the plant a good choice for the research of nematocidal activities.Rhizoma kaempferiae,a kind of perennial herb,the rhizome of which is massive and has aromatic odor.Rhizoma contains volatile components of Ethyl C innamic and is active in killing nematode,but it has not put into production and application yet.In order to apply Ethyl C innamic to prevention of nematodosis,the research adopt high-speed shear emulsification method and oscillating emulsification method to develop 10%Ethyl C innamic EW,and make evaluation on its effectiveness in killing nematode by pot experiment.The results are as follows:1.Use the method that combines high-speed shear emulsification and oscillating emulsion to select the auxiliary and the dosage o f the 10%Ethyl C innamate EW,and then obtain the optimal formulation of 10%Ethyl C innamate EW by orthogonal design method.The optimal formulation of Ethyl C innamate was 10%,xylene 5%,500#0.2%,TS-10 1.8%,NP-10P 0.5%,gum arabic 1.5%,deionized water to 100%.2.The quality of 10%Ethyl C innamate EW products was tested.The result showed that the content of Ethyl C innamate was 10.78%,and the emulsion?in 200 times solution?was qualified and stable with no sendiment,and the tipping stability was certified.Upon inspection,it separated no crystals out under cold storage and the decomposition rate under hot storage was 4.22%.The permanent foamability was 0.2cm and pH was 4.24.It has been proved that the emulsion conformed to national standard.3.The result of the toxicity measurement showed that the LC50 of 10%Ethyl C innamate EW to Bursaphelenchus xylophilus 2nd instar was 25.4?g/mL,and the effect was better than that of ethyl cinnamate original drug?32.25?g/mL?.4.The result of pot experiment showed that 10%Ethyl C innamate EW had better protective function on pine and controlled effect on pine wood nematode disease.The disease condition of 10%Ethyl C innamate EW?250?g/mL?developed slowly,the disease index was0.22 at 15 days after treatment,which was significantly lower than that of blank control?0.54?.After 15 days of inoculation,10%of the Ethyl C innamate EW-treated pine branches did not stop to flow of glue,while the CK of the pine flow was significantly reduced.
