Amino acids,it's the basic unit to constitute protein,is one of the essential nutrients to the human.This paper summarized the physiological functions and application fields of amino acids,and detailed introduction of lysine,arginine,leucine the three kinds of amino acids.Amino acid is widely used in the life,making the separation determination technology of amino acid is more and more important.This paper also described some common methods of separation and detection.By capillary electrophoresis method to detect the existence and content of material,it has good accuracy,high efficiency,good reproducibility and a series of advantages.Now has become a research focus in the field of analytical chemistry and is widely used in food detection.This paper mainly introduces the principle of capillary electrophoresis assay,detectable substances,and the method of use,also detailed introduced the basic concept of electrochemical luminescence.The purpose of this paper is to build a novel method for determination and separation of amino acids in beverages by capillary electrophoresis with an electrochemiluminescence detection?CE-ECL?based on tris?2,2'-bipyridyl?ruthenium?II?[Ru?bpy?32+].The main research contents and results are as fllows:1.Established a new method capillary electrophoresis separation-Ru?bpy?32+-electrochemiluminescence detection is applied to lysine testing in Red Bull vitamin drink.Through the experiments,the factors affecting the separation detection respectively are optimized.Under the best testing conditions,the ECL intensity of lysine and the concentration of lysine had a good linear relationship,its linear equation is Y=331.53+27.52x,linear range is 3.087.0?g/m L,the correlation coefficient is 0.9991,the lowest detection limit of lysine is 0.21?g/mL.The experimental results show that the content of lysine in Red Bull vitamin drink is 67.7?g/mL.2.Established a new method capillary electrophoresis separation-Ru?bpy?32+-electrochemiluminescence detection is applied to arginine testing in Ichimore drink.Through the experiments,the factors affecting the separation detection respectively are optimized.Under the best testing conditions,the ECL intensity of arginine and the concentration of arginine had a good linear relationship,its linear equation is Y=4.067x-10.432,linear range is 1.0440.22?g/mL,the correlation coefficient is 0.9995,the lowest detection limit of arginine is 0.24?g/mL.The experimental results show that the content of arginine in Ichimore drink is 34.96?g/mL.3.Established a new method capillary electrophoresis separation-Ru?bpy?32+-electrochemiluminescence detection is applied to leucine testing in Kuaibu amino acid drink.Through the experiments,the factors affecting the separation detection respectively are optimized.Under the best testing conditions,the ECL intensity of leucine and the concentration of leucine had a good linear relationship,its linear equation is Y=14.23x-1.21,linear range is 2.0320.0?g/mL,the correlation coefficient is 0.9998,the lowest detection limit of leucine is 0.13?g/mL.The experimental results show that the content of leucine in Kuaibu amino acid drink is 1054.8?g/mL.Compared with the national standard method,the CE-ECL method omits the pretreatment of complicated derivative,simplified the experimental steps to save time,improved the analysis efficiency,and had good reproducibility,low detection limit,is suitable for rapid detection of amino acids in beverages.
