Sugar and acid components,content and ratio are important indicators to measure fruit quality.Accurately determine the composition and content of sugar and acid in the fruit of Vaccinium uliginosum,and explored the excellent germplasm of high sugar and low acid.Collected and preserved,explored the correlation between the sugar content of the V.uliginosum and the geographical distribution and altitude,provided scientific basis for the further study of the Vuliginosum.Through high performance liqud chromatography and data analysis of 100 samples of V.uliginosum fruit from 10 different populations in Changbai Mountain,the results showed that:1.The organic acid chromatographic conditions were:Agilent Zorbax SB-Aq(4.6×250mm,5?m).The mobile phase was 0.01 mo 1/L K2HPO4(pH2.5)with a flow rate of 0.5 mL/min.The column temperature and UV detection wavelength were set as 30?and 210nm respectively(vitamin C 245nm),and sample size was 10?L.The sugar chromatographic conditions were:CAPCELL PAK NH2 UG80 S5(4.6×250 mm,5?m),mobile phase was acetonitrile:water(75:25),Evaporative Light Scattering Detector(ELSD),flow rate 1 mL/min;column temperature 35?;sample size was 5 ?L;drift tube temperature 80?,N2 for carrier gas,N2 flow rate 1 L/min;gain value of 1.2.Fructose and glucose were the main sugars in the V.uliginosum fruit,and their contents range from 18.60 to 38.54 mg·g-1FW and 15.77 to 35.08 mg·g-1FW,respectively.Quine acid,citric acid and malic acid were the main organic acid components,and their contents range from 16.06 to 20.97 mg·g-1FW,10.39 to 14.96 mg·g-1FW and 4.28 to 7.47 mg·g-1FW,Three of them account for 99%of total acid,oxalic acid and strontium.The content of oxalic acid,shikimic acid and vitamin C was low;the DFH ? population in the 10 populations had the highest fructose,glucose content and sugar-to-acid ratio,It is excellent populations.The total sugar content of MJ-7 plot is 92.40mg·g-1FW,the total acid content is 31.71mg·g-1FW,and the ratio of sugar to acid is 2.92,which is much higher than the other 99 plots.It is excellent in high sugar and low acid germplasm.3.Quinic acid and citric acid were significantly positively correlated with total acid,and fructose and glucose were significantly positively correlated with total sugar.Principal component analysis showed that oxalic acid,vitamin C,fructose,glucose and total sugar were the main components.The contribution is greater;quinic acid,citric acid and total acid contribute more to the main component 2.Cluster analysis showed that 10 populations could be divided into 4 categories based on sugar acid content.4.Fructose,glucose and total sugar content were positively correlated with altitude,negatively correlated with latitude,oxalic acid,vitamin C contents and sugar-acid ratio were positively correlated with altitude,organic acid and latitude were not correlated.Altitude and latitude were important environmental factors affecting the sugar and acid content of different populations of Changbai Mountain.
