Salvia miltiorrhiza Bunge,also known as the"red root",is a labiaceous plant which roots or stems can be used as medicines to treat cardiovascular and cerebrovascular diseases or to relieve the symptoms of oxidative aging.Studies have confirmed that the main medicinal ingredients of Salvia miltiorrhiza are diterpenoid quinones,phenolic acids,sugars,tanshinones,ketones,cactones,alkaloids and other chemical components.So far,about 100 chemical compounds have been discovered.Among them,lipid-soluble ingredients have good effects on slowing down cell oxidation,inhibiting cell carcinogenesis,as well as antibacterial activity and anti-inflammatory action.the water-soluble phenolic acids have the functions of cell protection,anti-blood clotting and anti-blood vessel thrombosis.In addition,Salvia miltiorrhiza contains many plant polysaccharide components with various physiological functions.Polysaccharide components can regulate the life activities of plants,It also has many applications in clinical medicine and health food industry.The water-soluble and lipid-soluble components of Salvia miltiorrhiza have been extracted and analyzed,and Salvia miltiorrhiza polysaccharide was systematically studied on its extraction,separation,purification and antioxidant activity.The research results are as follows:1.With the help of Response Surface Analysis Software,the optimum extraction process of effective components from Salvia miltiorrhiza was designed and verified.The optimal extraction process of lipid-soluble components was explored:the ultrasonic extraction temperature was 68 oC,the extracion time was 42 min,the ratio of material to liquid was 120 mL/g.The optimal extraction condition of water-soluble components was explored:the ultrasonic extraction temperature was 41 oC,the extraction time was 35 min,the ratio of material to liquid was 50 mL/g.2.The rhizomes of Salvia miltiorrhiza from Sichuan province?Zhongjiang County?,Anhui province?Bozhou City?,Gansu province?Qingyang City?,Hebei province?Nanhe County?and Shandong province?Xintai City?were selected in this study,the above optimal technological scheme was adopted to extract and analyze the differences of component contents in medicinal materials from different regions.The results showed that the compositions of Salvia miltiorrhiza are different in different places,so the content of active components of Salvia miltiorrhiza is closely related to its ecological environment.3.The content of salvianolic acid B in the water-soluble component of Salvia miltiorrhiza is low,but the content of the lipid-soluble component meet the standards of the“Chinese pharmacopoeia”.The contents of water-soluble components and lipid-soluble components of Salvia miltiorrhiza in Sichuan,Anhui,Hebei and Shandong provinces all meet the standards specified in“Chinese pharmacopoeia”.4.Salvia miltiorrhiza polysaccharide?SP?was prepared by Ultrasonic Method.SP0,SP1,SP2 and SP3 were separated and purified by using DEAE-52 cellulose column.In addition,their structure were characterized by FT-IR and UV.5.Different antioxidant evaluation systems were established to evaluate the antioxidant activity of Salvia miltiorrhiza polysaccharide?SPSP3?.The results showed that the scavenging ability of DPPH:SP>SP1>SP0>SP3>SP2,the hydroxyl radical scavenging ability:SP0>SP>SP1>SP2>SP3,the scavenging ability of O2-:SP>SP0>SP1>SP2>SP3,the scavenging ability of ABTS:SP>SP1>SP0>SP2>SP3.Therefore,Salvia miltiorrhiza polysaccharides?SP0SP3?have certain antioxidant activities.Among them,SP,SP0 and SP1 have great development and application prospects for pharmacy,because of their strong antioxidant activity.
