With the continuous progress of social economy,people are becoming increasingly aware of the importance of health.Nowadays,people's eating habits have changed a lot.The requirements of human being are no longer just living in the stage of adequate food,but also put forward for high-quality food in demand.Designing a fast and accurate food safety test system has become an important subject.Nano-probe technology which combining nanotechnology and biotechnology is a comprehensive interdisciplinary technology.Such technology is also a hot front-end technology in the field of international biotechnology research.In the medical and health,genetic engineering,microbial identification and other fields,the nano-probe technology has a very clear industrial background.Especially in the detection of medicine and health,the nano-probe technology can realize the specific identification and enrichment of bacteria,thus greatly improve the detection accuracy of bacteria detection.Graphene transparent electrode(GTE)materials have high transparency,light weight and good flexibility,and can be prepared in large quantities at low cost.Therefore,graphene transparent electrode are widely used in electronic equipment,optoelectronic equipment and energy storage equipment.In biochemistry,adenosine triphosphate(ATP)is a type of nucleotide.As the molecular currency of intracellular energy transfer,it is ubiquitous in all types of cells.ATP bioluminescence technique is a frontier method to detect the number of bacteria,the use of ATP is widespread,so special luminescence mechanisms can be used to detect the number of some microorganisms.This method greatly reduces the detection time which can obtain the results in twenty minutes,and has broad application prospects.This paper presents a biochemical system suitable for food safety evaluation.The technique of enrichment of bacteria with magnetic nano-probe and graphene transparent electrode is used in this system.The system applies ATP bioluminescence mechanism to detect food safety,it is more convenient than using traditional bacterial detection methods.The system not only uses the probe to capture and enrich Escherichia coli via an antibody-antigen reaction but also enriches ATP using an electric field generated by the GTE to further improve the accuracy of the system.This system can produce a linear correlation coefficient of up to 0.972 comparing with the other conventional approaches and meet the design demand.Moreover,detection can be completed within 20 min.The detectable bacterial concentration range is 10~2–10~6 CFU/mL.Additionally,a series of curves obtained by measurements of different polluted food products such as drinks,meat,and grains show that the system has a satisfactory performance.It meets the requirements of a rapid,on-site detection system.
