Agaricus bisporus is one of the largest mushroom species in the world.It is widely distributed in a wide range of areas.It is rich in phenolic substances.These phenolic substances have good anti-oxidation,anti-mutation,anti-tumor virus,antibacterial and slow osteoporosis,healthy teeth,blood lipids and blood sugar.Ultraviolet treatment has been widely used as a means of preservation,which not only can sterilize,but also increase the biologically active components in Agaricus bisporus.At present,there is no systematic study on the effect of ultraviolet light on phenolic substances in edible fungi.In this paper,white Agaricus bisporus is the main research object,and the ultrasonic extraction process of Agaricus bisporus is optimized to study the antioxidant activity and change of content of phenolic substances in Agaricus bisporus after UV irradiation.On the basis of single factor experiment,the ethanol volume fraction,liquid-to-liquid ratio,ultrasonic temperature and ultrasonic power were used to change the response value of Agaricus bisporus polyphenols.The optimal ultrasonic extraction conditions of Agaricus bisporus polyphenols were determined by response surface methodology.The results showed that with 70%ethanol as extract,the theoretical value of the extraction of polyphenols from Agaricus bisporus was 37.4876 mg/g under the conditions of ultrasonic temperature 65°C,ultrasonic time 60 min,liquid-to-liquid ratio 35:1 mL/g and ultrasonic power 400 W.The amount can reach 37.21 mg/g,the relative error is 0.74%.The optimal process conditions for response surface methodology optimization can be applied to the extraction of polyphenols from Agaricus bisporus.Determination of phenolic substances in Agaricus bisporus by high performance liquid chromatography-mass spectrometry,optimized conditions:ZORBAX Edipse XDB-C18Rapid Resdution(4.6×150 mm,3.5?m)column;0.1%(v:v)formic acid Aqueous solution-acetonitrile,elution gradient 0~5 min,10%B;5~12 min,17%B;12~25 min,17%B;25~30 min,50%;30~35 min,70%35~40 min,10%;flow rate 0.4 mL/min;injection volume 10?L,column temperature 30°C.In the ESI-ion scan mode,the linear curves of the seven standard standards are good,the correlation coefficients are all greater than 0.999,the limit of quantitation is low,and the precision is high.The relative standard deviation(RSD)of the six parallel determination standards is less than 5%;The recoveries of horizontal spikes ranged from 89.36%to 99.07%,and the relative standard deviations(RSD)ranged from 0.39%to 5.87%,indicating that the established instrument conditions meet the experimental requirements and can be used for separation and detection of bisporus.Using 2 kJ/m~2 irradiation dose,firstly,different kinds of ultraviolet radiation were applied to Agaricus bisporus,followed by ultraviolet irradiation of Lentinus edodes and Pleurotus ostreatus.For further exploration,UV-C was used to treat intact complete bisporus,Agaricus bisporus pileus,Agaricus bisporus gill,and Agaricus bisporus stipe.The cover and the whole mushroom,the mushroom stipe,and the mushroom cover were irradiated,and compared with the non-UV-C irradiation,And determine the antioxidant activity of Agaricus bisporus and its various parts.The results showed that the content of phenolic substances in Agaricus bisporus increased after UV irradiation.Taking chlorogenic acid in the phenolic substance of Agaricus bisporus as an example,the chlorogenic acid increased by 1.33 times after UV-B irradiation,and UV-C irradiation increased by 1.17 times,using UV-C and UV-B mixed irradiation,UV-B irradiation followed by UV-C irradiation,and UV-C irradiation and UV-B irradiation,the difference in irradiation order was not obvious;The content of phenolic substances in different mushrooms,Gray oyster mushroom>Mushroom>Agaricus bisporus.Unirradiated Agaricus,gill>stipe>complete bisporus>pileus.After UV-C irradiation,the content of each part of phenolic substances was increased.Antioxidant with VC and BHT as controls,the Agaricus bisporus,Agaricus bisporus gill,Agaricus bisporus stipe,Agaricus bisporus pileus and UV-C irradiated Agaricus bisporus,Agaricus bisporus gill,Agaricus bisporus stipe,Agaricus bisporus pileus polyphenols were studied.Reducing power,hydroxyl radical scavenging ability,DPPH free radical scavenging rate,superoxide anion scavenging ability.The experiment shows that the extract of polyphenol extract of Agaricus bisporus has low IC50 value,good scavenging ability to hydroxyl radical,DPPH free radical and superoxide anion,and its free radical scavenging ability after UV irradiation increases,showing stronger antioxidant activity.Ability,so ultraviolet irradiation of Agaricus bisporus will increase the phenolic substances in Agaricus bisporus,thereby improving its antioxidant capacity.
