This experiment was mainly regarded polysaccharides of racilaria lemaneiformis as the research object.The research analyzed its compositions and structure and studied its antitumor mechanism in vivo and in vitor.At last the experiment investigated the immunologic mechanism of immune responses in the body.This research subject had importance for tumor immunotherapy and improving the bioavailability of racilaria lemaneiformis polysaccharides.Chemical analysis and the FT-IP spectra of racilaria lemaneiformis polysaccharides showed that it was a typical acidic polysaccharide which has high content of uronic acid and sulfo group.The Uv-vis spectra of the polysaccharides showed a strong characteristic absorption of polysaccharides and did not have absorptions of nucleic acids or proteins.The GC analysis showed that these polysaccharides were mainly composed of galactose,whose percentage was 60.43%.Firstly,the research focused on antitumor activity of the polysaccharides form racilaria lemaneiformis in vivo.Five tumor bearing mice could not be checked out in each group by the polysaccharides with intragastric dose of 250 mg/kg·d.It was indicated that the polysaccharides could effectively inhibit the proliferation of solid tumor.By measuring the immune organ index of mice,we found the polysaccharides had a very good protection effect on tumor-burdened immune organs in mice.The effects of polysaccharides on the index test of thymus and spleen were studied in tumor-bearing mice.It was found that the polysaccharides could effectively portect the thymus and could promote the normal development of T lymphocytes.The polysaccharides could strengthen proliferation activity of T lymphocytes and B lymphocytes by testing the spleen lymphocyte proliferation ability.The NK cells killing the target cells ability and the proportion of T lymphocytes in the spleen had significantly enhanced in racilaria lemaneiformis polysaccharide group.The indicators of peripheral blood test showed that the polysaccharides could balance the distribution of blood cells in tumor-bearing mice.The polysaccharides could enhance the activity of T cells and significantly enhance the activity of B lymphocytes to generate a large number of specific antibody IgG levels.It was showed that the polysaccharides can improve the level of IL-2?IFN-??TNF-? in the serum of tumor-burdened mice.The polysaccharides could make tumor regression by restaining the tumor cells in G0/G1 phase and inducing the tumor cells apoptosis through solid tumor index test.Secondly,it was studied that the in vitro antitumor activity of racilaria lemaneiformis polysaccharides towards S180 sarcoma cells with different concentrations and treatment time.It was found that there were significant inhibition and apoptosis induced by racilaria lemaneiformis polysaccharides.Co-cultured with 200 ?g/mL polysaccharides for 72 h,the highest inhibition ratio toward S180 cells was 55.54%.These demonstrated that the polysaccharides have significant antitumor activity in vitro.
