Nasopharyngeal cancer(NPC)is a malignant tumor of the head and neck,which often occurs in the mucous membrane of the rhinitis.It has a high incidence and mortality in the southern provinces of China.As a molecular fingerprint spectrum,Raman spectroscopy is capable of detecting the characteristic structure of biomolecules,which is suitable for rapid and non-invasive detection of biological sample.Surface-enhanced Raman spectroscopy(SERS)technology can enhance the weak Raman signal by 1013-1015 times,and has the advadtages in high detection sensitivity and multiplex detection capability.This thesis will apply above two techniques for the analysis of blood,ex vivo tissue and in vivo tissue of nasopharyngeal cancer patients.Meanwhile PCA-LDA multivariate statistical algorithm is employed for the analysis of the collected spectra,with the aim of developing a fast,objective and accurate detection method for nasopharyngeal cancer.The main research results are as follows:1?A total of 50 nasopharyngeal cancer serum samples and 50 normal ones were tested by the 633nm and 785nm laser,respectively.The diagnostic sensitivity of 100%and 100%can be achieved by the two kinds of excitation light,and corresponding specificity are 96.5%and 100%.This shows that the diagnostic efficiency under the excitation of 785 nm wavelength is slightly better than that under the excitation of 633 nm wavelength.2?Silver nanoparticle based surface-enhanced Raman spectroscopy was applied to analyze nasopharyngeal caner and non-cancer tissue.Using PCA-LDA multivariate analysis,we were able to differentiate NPC tissue from non-cancerous tissue with the diagnostic sensitivity of 78.3%and specificity of 84.3%.Some changes of specific biochemical components in cancer tissue samples were found by analyzing the changes from SERS peak intensity.For example,the levels of phenylalanine,DNA and protein in cancer tissues increased in comparison to nasopharyngeal non-cancerous tissue.Results showed that nanoparticle based SERS technology combined with PCA-LDA has potential to be an objective,rapid and accurate method for cancer detection.3?White light endoscope-guided nasopharyngeal tissue detection system wras employed to study the NPC in vivo tissue.Comparing the attribution of Raman peaks in cancerous and non-cancerous tissues and the differences in intensity,we found that the content of various biological substances in cancerous tissues changed,such as a significant increase in the content of nucleic acid.The results show that Raman spectroscopy combined with multivariate statistical analysis methods has high detection sensitivity and specificity for NPC screening.
