Arsenic is recognized as the toxic and hazardous substances because of their potential risks to human health,it is susceptible to be absorbed by animals and plants through the food chain.Whie speciations of arsenic are closely related to the bioavailability and the toxicity.In recent years,a number of analytical techniques have been employed for speciation analysis of arsenic.To our best knowledge,there are few reported works for analysis of arsenic speciation of edible fungus and the changes of concentration,speciation and bioavailability of arsenic after cooking.An analysis method of high performance liquid chromatography with inductively coupled plasma mass spectrometry for arsenic speciation analysis was described.On this basis,detect the changes of various arsenics by vitro test under the cooking conditions,the bioavailability and toxicity of the samples were evaluated,and the contect of arsenic and arsenic speciation in edible fungus samples were determined,The primary research result involves:1.An analysis method of HPLC-ICP-MS for arsenic analysis was established.The species includes As?,As?,MMA,DMA,AsC,AsB which were well separated in 15min.The effect of composition,concentrations,pH and flow rate balance of the mobile phase on separation of six arsenic species has been investigated.Under the optimized conditions,there were good linear relationships in 0-50p.g/L for six arsenic compounders,the correlation coefficients were up to 0.9988.The detection limits of the species studies were AsC 0.31?g/L,AsB 0.51?g/L,As?0.39 ?g/L,DMA 0.37?g/L,MMA 0.46 ?g/L,Asv 0.59?g/L.All the recoveries were better than 94.30%.The relative standard deciation(RSD)was less than 5%.This method was simple,rapid,sensitive and accutate,met the arsenic speciation quantitative requirements.2.The analysis of arsenic species in dried L.edodes samples,raw L.edodes samples and L.edodes chips samples.The contents of total As in samples were determined by inductively coupled plasma mass spectrometry(ICP-MS)system.Meanwhile,This paper evaluates the effect of ultrasonic conditions on the stability of arsenic compounds,and on this basis,a sequential As ultrasonic assisted extraction method of dired L.edodes samples were tested,Subsequently,0.3 mol/L acetic acid ultrasonic-assisted extraction(300 W,10 min,60?)was chosen.About 76.84%As species compounds could be extracted.The dominant As species identified in L.edodes samples were inorganic arsenic(iAs),inorganic arsenic accounted for 50%of total arsenic.3.Analysis of the effect of microwave treatment on the stability of the arsenic compounds,the results show that arsenic compounds were relatively stable when measured under microwave treatment conditions(microwave power is 150 W,300 W and 450 W and microwave time is 30-150 s).And on the basis,the research evaluates the effect of microwave blanching(MB)on the content of total arsenic and arsenic compounds in raw L.edodes samples,the results showed that the content of total arsenic and iAs in all tested samples after MB treatments were released and reduced to about 9.14-37.09%of those raw mushroom.Inorganic arsenic(iAs)was found to be the predominant arsenic species in L.edodes before and after MB.Furthermore,arsenic species in raw mushroom and the MB treated(300 W,90 s)mushroom were determined in the gastric and gastrointestinal bioaccessible fractions obtained after the simulated in vitro gastrointestinal digestion.The dominant As species identified in L.edodes samples were As?,and the As? accounted for the proportion of the total arsenic is bigger,the two samples after the simulated in vitro gastrointestinal digestion,the proportion of As? was declined,while the proportion of Asv was increased slightly,As?concentrations in samples which were subjected to microwave blanching was reduced and As? concentrations was increased.Results show that although the ratio total inorganic arsenic to total arsenic in sample change was small after microwave blanching,because the As? toxicity is higher than As?,so microwave blanching can reduce the consumption of risk of edible fungi.4.The analysis of As species in L.edodes samples.The total arsenic in dried L.edodes samples,raw L.edodes samples and L.edodes chips samples were determined by ICP-MS.Concentrations of arsenic in all tested samples were 0.1969?0.8171 mg/kg,0.2429?1.9714 mg/kg,0.0306?0.2269 mg/kg,respectively.Finally,the concentration of As in one batch of raw L.edodes samples was over standard,no As in dried L.edodes samples and L.edodes chips samples was over standard.The dominant species identified in L.edodes samples were inorganic arsenic,inorganic arsenic accounted for 50%of total arsenic.AsC was only detected in dried L.edodes samples(Qingchuan county,sichuan province),MMA was detected in all raw L.edodes samples except the first sample,and no As? was dected in L.edodes chips(Zhangzhou in fujian province).
