Pomegranate(Punica granatum L.)deciduous tree or shrub,dicotyledon,which belongs to the genus Punica in the family Punicaceae.China's pomegranate is rich in resources,pomegranate is usually only eaten as a fruit,and most of the pomegranate leaves are discarded.Pomegranate leaves contain a variety of active ingredients,such as tannins,flavonoids,alkaloids,etc.Therefore,exploring the potential value of pomegranate leaves is of great significance to the sustainable and healthy development of the pomegranate industry.In this study,pomegranate leaf were used as raw materials to process conditions of flash extraction of polyphenols from pomegranate leaf were optimized,and the purification,activity and application of polyphenols from pomegranate leaf were studied.The main contents are as follows,(1)Optimization of flash extraction of polyphenols from pomegranate leafTaking the extraction amount of crude polyphenols from pomegranate leaves as the index,the ethanol concentration,the ratio of material to liquid and the extraction time were selected as the investigation factors.Based on the results of single factor experiment,the response surface experiment was designed.The optimal process conditions for the flash extraction were obtained.The ethanol concentration was 57.56%,the solid-liquid ratio was 1:8.65(g/mL),the extraction time was 3.95 min,and the theoretical predicted extraction amount was 29.37 gGAE/100 g.Considering the actual operation,the flashing conditions were adjusted to,ethanol concentration was 58%,solid-liquid ratio was 1:9(g/mL),extraction time was 4 min,and the verification result was 29.12 gGAE/100 g,which is close to the theoretical prediction value.(2)Purification and activity of polyphenols from pomegranate leafPurification of polyphenols from pomegranate leaf with AB-8 macroporous resin,adsorption performance and desorption of AB-8 macroporous resin at 25 ?,sample concentration controlled at 0.9947 mg/mL,ethanol concentration of eluent 70% The performance was the best,the adsorption rate was 90.72%,and the desorption rate was 72.3%.VC and gallic acid were selected as the control.The reducing power of polyphenols from pomegranate leaf,the scavenging effect on DPPH free radicals and hydroxyl radicals and the inhibitory effect on tyrosinase activity were investigated.The results showed that polyphenols from pomegranate leaf had good antioxidant activity and inhibition of tyrosinase activity.(3)Formulation design and evaluation of O/W anti-aging emulsion with polyphenols from pomegranate leafAccording to the characteristics of the emulsion cosmetics and the design principles of the anti-aging emulsion,based on the preliminary pre-test,the composition and dosage range of the formulation are formulated.The best basic formulas obtained by single factor experiments and orthogonal tests were: glycerol(4%),1,3-butanediol(3%),isopropyl palmitate(4%),squalane(3%),Lanolin(3%),VE(1.5%),D-panthenol(3%),polyphenols from pomegranate leaf(1.5%).A complex emulsifier of A165(2%)and PGFE(1%),a complex thickener of PEG-120(0.05%)and xanthan gum(0.3%)can be selected to stabilize the product quality.According to the emulsification technology and the emulsification principle,under the optimal formulation conditions,the optimal emulsification conditions are emulsification temperature 85?,emulsification time 20 min,emulsification speed 50 r/min.The sensory evaluation index of the self-made anti-aging emulsion is superior,and the microbial and heavy metal indexes meet the requirements of the light industry industry standard.The self-made anti-aging emulsion containing polyphenols from pomegranate leaf has a certain anti-oxidation property compared with the commercially available anti-wrinkle firming anti-aging emulsion containing the compound extract and the water-coded red pomegranate series.Therefore,polyphenols from pomegranate leaf can be used as a natural antioxidant in combination with other antioxidants and applied to the cosmetics industry.
