Modern research shows fruits and vegetables are rich in polyphenols、flavonoids and other active substances. Polyphenols in fruits and vegetables have significant antioxidant activity, they can remove the free radicals, and also have proliferation inhibition effects on some tumor cells. Ipomoea batatas leaves, as a kind of edible efficacy of vegetable, is widely grown in China and it has good prospects of development. So, in this paper, we put 11 different varieties of Ipomoea batatas leaves in Guangdong as our research samples, to explore the polyphenol and the contents of the functional components extract in Ipomoea batatas leaves, then determine the extracellular and intracellular antioxidant activity of the Ipomoea batatas leaves extract, to study their inhibition of cytotoxicity and proliferation of different tumor.In this study, different varieties of Ipomoea batatas leaves are the samples. We take different extraction methods to extract the free and bound polyphenols in the Ipomoea batatas leaves samples, then measure the contents of polyphenols and flavonoids. From the results, we find that polyphenol contents among different varieties of Ipomoea batatas leaves are obvious differences and the vegetable type of Ipomoea batatas leaves are not much advantage. Different varieties of Ipomoea batatas leave have little flavonoid content, the vegetable type of Ipomoea batatas leaves have high content. Overall, the Ipomoea batatas leaves named Guangcai 4 better than other Ipomoea batatas leaves.We measure the active ingredient in the Ipomoea batatas leaves by HPLC. We take the preliminary high-performance liquid chromatography scan and analysis of different varieties of Ipomoea batatas leaves samples, the results show that HPLC spectrums of Ipomoea batatas leaves are similar, which means they contain the same characteristic components. The peak time of Ipomoea batatas leaves is 13 min. In the HPLC spectra of the Ipomoea batatas leaves, the peaks’ shapes are complex. All these indirectly proof that Ipomoea batatas leaves are rich in the active ingredients.Use Guangcai 4 as the sample, we take 4 different ways to test the antioxidant activity of Ipomoea batatas leaves extracts in vitro. The extracellular antioxidant activity assay results show that ethyl acetate extracts were best antioxidant in vitro. The intracellular antioxidant activity assay results show that the CAA in Chloroform extract is the highest, which is 169.96μmol QE/100 g fresh weight(PBS wash) and the CAA in ethyl acetate extract is the highest, which is 1673.10μmol QE/100 g fresh weight(no PBS wash).Use Guangcai 4 as the sample, we research Ipomoea batatas leaves extracts cytotoxic and anti-proliferative effect to Hep G2 and MDA-MB-231. We found that Ipomoea batatas leaves extract have more significant inhibition of proliferation. To Hep G2 cells, Ipomoea batatas leaves extracts have significant proliferation inhibition effect. To MDA-MB-231 cells, inhibit the proliferation of cell lines in Ipomoea batatas leaves extracts have differences. The ethyl acetate extract has the strongest ability of anti-proliferative effect.
