Cow's milk has gradually become the main food protein source for infants.However,cow's milk is also one of the eight major allergens confirmed by the World Allergy Organization(WAO).In recent years,with the the deterioration of the environment,cow's milk allergy for infant is becoming more and more serious.It is reported that the incidence of cow's milk allergy in preschool children ranges from 1% to 17.5%.There are more than 30 kinds of proteins in milk,all of which are potential allergens,among which the main allergens are casein,?-lactalbumin and ?-lactoglobulin.The enzymatic hydrolysis method can effectively degrade these proteins.Moreover,the condition is mild and the process is simple,which has incomparable advantages compared with other methods.It is the main method for the production and development of hypoallergenic infant formula.At present,most of the hypoallergenic milk powder in the domestic market is imported from abroad or produced under original equipment manufacturer,and there is no such hypoallergenic milk powder in China.Therefore,it is urgent to develop the hypoallergenic milk powder for demostic allergic children.The research contents of this paper include: the establishment of the Complex enzymatic hydrolysis scheme of whey protein,the preparation of hypoallergenic infant formula and evaluation of its allergenicity,and plant design of the production of hypoallergenic infant formula.The research methods,results and conclusions are as follows:1.Based on the ratio of enzyme to substrate,the time,the initial pH value and the temperature of enzymatic hydrolysis,the conditions for the enzymatic hydrolysis of whey by three proteases were optimized,and then the complex enzymatic hydrolysis of whey was conducted on this basis.From the result,the optimal hydrolysis conditions is that whey protein were hydrolyzed by trypsin firstly at 40.63 ?and pH 7.37 with E/S of 4% for 3 h,then the reaction were adjusted to 52.3 ? and pH 7.2 with E/S of 4% by compound flavor protease for 3 h.2.According to the national standard and the ingredients of breast milk,the hypoallergenic formula was prepared with the hydrolyzed whey protein.The flavor of the milk powder was greatly improved and the bitterness was not obvious compared with the enzyme-hydrolyzed whey.3.With sera from patients allergric to milk,specific IgG/IgE binfing ability to the main allergens in whey powder has decreased significantly,however,the binding ability of hypoallergenic infant formula has increased,but still lower than that of commercial milk.Moreover,the binding ability of specific antibody to the two allergens was further compared,and it was found that ?-lactoglobulin by the complex enzyme was degraded more extensively than ?-lactoalbumin.4.In vitro experiment results of KU812 cell model,it showed that the release capacity of histamine,?-hexoaminase and IL-6 stimulated by enzymatic hydrolysis of whey protein decreased with varying degrees,among which histamine showed the most obvious changes.Compared with the commercial hypoallergenic milk powder,the release capacity of ?-hexoaminase in the homemade hypoallergenic milk powder was 16.788% lower,the release volume of IL-6 was lower by 28.71pg/mL,and the release volume of histamine was lower by1.11ng/mL.5.Balb/c mice test results showed that: the specific antibodies IgE/IgG and histamine of mice from different groups were determined,proving that the sensitization model by Balb/c mice was successfully constructed;The allergeniciy of different samples was evaluated by detecting T cell differentiation and cytokine levels.The change trend of its allergenicity was from low to high as followes:for the commercial hyposensitized milk powder(deep hydrolysis)< enzymatic hydrolysis whey < homemade hypoallergicgenic milk powder < commercial ordinary milk powder.6.Based on small scale laboratory experiments with hypoallergenic milk powder,and the factory design was carried out.The factory layout,equipment selection and related drawings of hypoallergenic milk powder were performed.The designe scale is with annual output of 250 tons of low-sensitization milk powder,annual sales of 100 million yuan and annual net profit of 35.3551 million yuan.
