Pterostilbene(PT)is a natural active compound derived from plants such as red sandalwood,blueberry,grape,etc..It can improve blood cholesterol levels,prevent diabetes,fight cancer,fight inflammation,resist oxidation,relieve pain,treat and protect skin diseases.The water solubility of red sandalwood is poor,resulting in low bioavailability,which is limited in application.In this paper,the high-pressure homogenization method was used to prepare the pterostilbene nanoemulsion.Six single factors were selected,which were oil phase,emulsifier,emulsifier addition,oil phase to water phase ratio,high pressure homogenization pressure and homogenization cycle.The results showed that linseed oil was used as the oil phase,Tween 80 was used as the emulsifier,the emulsifier was added 1.5 times as much as the oil,the water phase was 85%compared with the oil phase 15%,and the high pressure homogenizing pressure was 800 bar,5 cycles are the best result of single factor.This thesis further explored the analysis of fatty acid composition and intestinal flora in healthy mice by pterostilbene nanoemulsion.The fatty acid composition of the tissue was determined by gas chromatography,and the composition of the fecal flora was determined by 16S rRNA high-throughput sequencing technology.A total of 42 healthy 6-week-old male C57BL/6 mice were randomly divided into 6 groups:blank group(C),oil group(O),pterostilbene group(PT),pterostilbene oil solution group(O.PT),Low-concentration emulsifier nanoemulsion group(LE.NE,emulsifier content 12.5%)and high-concentration emulsifier nanoemulsion group(HE.NE,emulsifier content 25%),The main test results are as follows:1.Under the intervention of the pterostilbene nanoemulsion,the low concentration emulsifier pterostilbene nanoemulsion group and the body weight of mice in the high-concentration emulsifier nanoemulsion group was also significantly lower than that in the pterostilbene oil solution group.2.In the serum of mice,the concentration of total cholesterol,high-density lipoprotein cholesterol and low-density lipoprotein cholesterol in the pterostilbene oil solution group was lower than that in the oil group.After the pterostilbene was embedded as nanoemulsion,the concentrations of total cholesterol,high-density lipoprotein cholesterol and low-density lipoprotein cholesterol were significantly lower than those of the pterostilbene oil solution group.As the concentration of emulsifier increased,the concentration of total cholesterol decreased significantly,but the concentration of low-density lipoprotein cholesterol increased.3.In the mice liver,also,the concentration of total cholesterol and low-density lipoprotein cholesterol in the pterostilbene oil solution group was significantly lower than that in the oil group.Under the intervention of pterostilbene nanoemulsion,the total cholesterol concentration of the high concentration emulsifier nanoemulsion group was significantly higher than that of the pterostilbene oil solution group,and the triglyceride concentration was significantly decreased.4.The results of fatty acid distribution in mice showed that the content of polyunsaturated fatty acids detected in the serum,liver and small intestine of mice was affected by the pterostilbene.The content of various polyunsaturated fatty acids in the low-concentration emulsifier nanoemulsion group was significantly higher than that in the pterostilbene oil solution group.The content of various polyunsaturated fatty acids in the high concentration emulsifier nanoemulsion group was significantly lower than that in the low concentration emulsifier nanoemulsion group.5.The effect of pterostilbene on the intestinal flora of healthy mice showed that pterostilbene significantly increased the microbial diversity in the intestine of mice and changed the structure of the flora.In the microbial community composition,the relative abundance of the Firmicutes in the pterostilbene oil solution group decreased,and the relative abundance of Bacteroidetes increased.However,the relative abundance of the Firmicutes increased and the relative abundance of Bacteroidetes decreased compared with the pterostilbene oil solution group.The pterostilbene nanoemulsion is made of oil-soluble type of nanoemulsion to improve the solubility of pterostilbene,improve its bioavailability and increase its stability.At present,the research on the preparation process of pterostilbene nanoemulsion in China is less effective.It provides an effective method for the better utilization of pterostilbene nanoemulsion in the food field.The effect of pterostilbene on tissue fatty acid metabolism provides a reference for studying the metabolic fate of long-chain polyunsaturated fatty acids in vivo,and the effect on the intestinal flora of mice may reveal the health effects of pterostilbene.
