Emamectin benzoate has strong insecticidal activity against Bursaphelenchus xylophilus.The formulation of emamectin benzoate soluble granule for trunk injection is in order to prevent and control pine wilt disease effectively.Basing on the formulation and production process of emamectin benzoate soluble granule,the conduction efficiency of soluble granule in Pinus massoniana is determined and the distribution of emamectin benzoate in the structure of P.massoniana is characterized in situ.After trunk injection,we obtain the control effect of the pine wood nematode disease and the residual distribution dynamics of emamectin benzoate in P.massoniana.The main findings of this paper are as follows:1.The optimum formulation and granulation process was studied.The solubility and oil absorption rate of six water-soluble carriers were determined,then we used the uniform design of experiment with mixtures and partial least squares regression to acquire the optimum assembly,stirred well with the emamectin benzoate oil phase,dried and squeezing granulation.According to the lab procedure,The pilot process used ZGH high-speed kneader to knead the material,determined the optimum kneading time was 10 min.Finally,the SET-100 single-screw extruder was used for extrusion granulation,we obtained optimum formulation emamectin benzoate 14.3%?pure 10.0%?,surfactant 5.7%,cosolvent12%,sodium benzoate 39.0%,lactose monohydrate 20.5%,water-solubility starch 3.0%,CMC 4.5%,and the properties of the granule were in line with the requirements..2.The conduction efficiency of granule was studied and the distribution of emamectin benzoate in P.massoniana was characterized.Segmented a sample of P.massoniana after 30 days for trunk injection,crushed and HPLC for determination.The content of emamectin benzoate could be detected in all parts,the effective conductivity of emamectin benzoate was during 17.38%and 28.67%in 30days test.Repeated the same operation after 60days,we obtained the effective conductivity of emamectin benzoate was during 49.08%and 70.30%.The main stem of P.massoniana was derivatived by fluorescence,and the sections were observed by fluorescence microscope.It was found emamectin benzoate was mainly distributed in the xylem axial resin channel,the tracheid and the sapwood part of the xylem.The result showed that granules can be dissolved and effectively distributed in P.massoniana.AfterBursaphelenchus xylophilus invaded the pine tree,it firstly fed on the parenchyma cells,and the emamectin benzoate could kill Bursaphelenchus xylophilus and then prevented the propagation and spread of Bursaphelenchus xylophilus in P.massoniana.3.The residual dynamics and control effect of pine wilt disease after trunk injection in forest were studied.The results of the sampling test were 0.97 mg/L,1.56 mg/L for the 60 day and 180 day after trunk injection in Xian'gong Mountains,and the Donghu Village's residuals were 1.627 mg/L and5.484 mg/L,which were higher than the pine wood nematode LC90?0.75mg/L?.In the pine wood nematode disease area of Donghu Village,110 pines were injected,the mortality of P.massoniana was 0%after 1 year,and meanwhile the mortality of P.massoniana in control plot was 6.1%.In the Xian'gong Mountain,230 pines were injected,the mortality of P.massoniana was 1.7%,and the mortality of P.massoniana in control plot was 10.8%.The emamectin benzoate soluble granule has the characteristics of simple and safe production,no plastic bottle package will not cause waste of plastic container carrier and environmental pollution and significant control effect.It has a good application prospect in the prevention and control of pests and diseases in forests and urban greening trees.
