Nano-silver?AgNPs?is one of the most widely used nano-metal materials.The previous reports have shown that AgNPs has obvious toxic effects on organisms.However,studies on the effects of nanosilver on the reproductive system are rare,and the direct toxicity of AgNPs to human embryos,especially to embryos,is still unclear.In this study,we used mouse embryos to study the developmental toxicity of metal nanomaterials AgNPs on preimplantation embryos in vitro.In order to clarify the direct effect of AgNPs on mammalian embryos and related pathways,this study used mouse preimplantation embryos as materials to establish an in vitro model of nanosilver embryo toxicity using IVF technology.To analyze the developmental pattern of preimplantation embryos cultured in culture medium containing AgNPs with different particle sizes and concentrations,and the time effect of nanosilver on developmental toxicity of preimplantation embryos.To determine the link between embryonic developmental toxicity and nanosilver.In this study,immunofluorescence technique,DCFH-DA,DiBAC4?3?,JC-1 and other tests were used to detect ROS levels,mitochondrial damage and membrane potential changes in embryonic cells under different environmental conditions,and the activity changes of Casepase-3 in early embryos were studied.The results showed that the toxic effect of AgNPs on embryonic development showed particle size-concentration-time dependence;AgNPs treatment of preimplantation embryos disrupted the normal formation of blastocyst lineage,and the expression balance of lineage markers Cdx2and Oct4 occurred.Alterations cause abnormalities in embryonic developmental fate;AgNPs in culture systems lead to elevated levels of ROS in the embryo and impaired mitochondria;nanosilver causes changes in membrane potential of embryonic cells;nanosilver causes changes in Caspase-3 activity in early embryos.AgNPs impair the development of mouse embryos to a large extent,and induce damage to embryonic development.The effect of AgNPs on embryos was analyzed.In conclusion,there are two pathways for cell death caused by AgNPs.Pathway 1:AgNPs causes toxic effects in embryonic development due to elevated ROS levels in embryos.Pathway 2:AgNPs changes embryonic membrane potential And increase the number of embryonic cells in the embryo and induce abnormal development and apoptosis of embryonic cells.
