Corneal contact lens,also known as contact Lens,is a lens made of transparent or dyed material based on the shape of a human cornea,it is directly attached to the tear layer of cornea and can be physically compatible with the human eye,and ultimately achieve the purpose of corrected vision,beauty or treatment.During the wearing process,corneal contact lens absorbs proteins,lipids,collagen,etc.in the tear fluid,which makes it easy to breed pathogens such as Staphylococcus aureus and Aspergillus,in addition,side effects such as cornea edema,cornea neovascularization,and allergic reactions can occur,and eye diseases such as cornea inflammation and conjunctivitis can be caused.If the antibacterial lens can be prepared and the bacteria growth on its surface can be inhibited directly,the phenomenon of bacteria or other microbes can be overcome fundamentally on the soft mirror surface.In view of the above background,this paper revolves around the preparation of antibacterial corneal contact lens and conducts research in three aspects:antibacterial agent preparation,antimicrobial evaluation and biocompatibility.The specific research work is as follows:1)Two antimicrobial agents containing quaternary ammonium salt functional groups were prepared.The antibacterial agent 1 was obtained by heating and stirring refluxing of N,N-dimethyl-Aminopropyl group-Trimethoxysilane and N,N-Dimethyl butylamine,and which the yield was 85%;The antibacterial agent 2 was prepared by refluxing Bromohexane with Dimethylamino ethyl methacrylate,which as a raw material,with a yield of 50%.The structure information of the two antimicrobial agents was confirmed by NMR and IR spectra.Next,the surface quality,transmittance,residual amount,elongation,contact angle and other parameters of the antibacterial corneal contact lens produced by the covalent grafting method and crosslinking grafting method were evaluated.Among them,the antibacterial corneal contact lens made by covalent grafting has the problems of stickiness and serious ester absorption.The corneal contact lens experimental results obtained by crosslinking grafting show that when the blending amount was 1%,the obtained soft mirrors can reach the standard from the measured parameters such as transmittance,contact angle,and elongation.2)Antibacterial corneal contact lens was prepared and its antibacterial properties were tested.First of all,the antibacterial properties of the antibacterial agent itself were evaluated,followed by exploration of the Oscillatory flask method and improvement of the"QB/T 2591-2003 Test Method for Antibacterial Properties of Antibacterial Plastics."Experimental results showed:Antibacterial agent 1 and antibacterial agent 2 have good bactericidal effect on Bacillus coli,and antibacterial agent 2 has better antibacterial performance;Oscillatory flask experiment overlap can not simulate real wearing condition;By testing the antibacterial performance of the antibacterial corneal contact lens on Bacillus coli,it was found that the antibacterial rate could reach 80%or more for the bacterial liquid with a concentration of 106.Compared to blank lenses,1%of antimicrobial lenses can effectively inhibit the growth of Staphylococcus aureus;the antibacterial effect of 5%antibacterial lens on albicans saccharomyces was obvious after more than 5 hours,and the antibacterial rate could reach 80%.3)A series of biocompatibility tests were carried out using the prepared corneal contact lens.According to?GB/T 16886.5-2003 Biological evaluation of medical devices?the extraction solution experiments showed that the relative proliferation rate of 0.5%antibacterial soft lens was 47.7%.When the cross-linking agent was doubled,the relative proliferation rate of 0.5%antibacterial soft lens increased from 50%to about 90%.Referring to the national standard,0.5%of the antibacterial soft mirror obtained by agar diffusion method basically reached the first class standard.
