Oat bran and 14 kinds of mushrooms were studied in this thesis.A method for the determination of antioxidant content in oats and mushrooms was established.Extraction,purification and antioxidant activity analysis of avenanthramides were carried out.The mushroom extracts were prepared,and their antioxidant content and antioxidant activities were analyzed.Oat and mushroom antioxidant extracts were applied to cosmetics to prepare a skin care lotion,and their physical and chemical properties,stability and antioxidant activities were evaluated.Main contents and results are as follows:1?An acid dye colorimetric method was established to determine the content of total alkaloid in oat with absorbance as the index.The optimal test condition is: the acid dye is bromocresol purple,the buffer is sodium dihydrogen phosphate-sodium hydroxide at pH 5.4,and the acid dye dosage is 3 mL;A HPLC method for the determination of glutathione and ergothione in mushrooms and a method for the determination of total polyphenols by uv-visible spectrophotometry were established.2?The extraction and purification process of avenanthramides were studied with avenanthramides yield(mg/kg)as an index.The optimum extraction condition of oat alkaloids by flash type high speed extractor was as follows: liquid-solid ratio 19:1(mL/g),80% ethanol concentration,extraction temperature 73?.The optimal conditions for the purification of avenanthramides were: D-101 macroporous resin was selected,the sample concentration was 34.7 mg/L,the concentration of ethanol elution was 90%,the flow rate of the sample was 1.0 mL/min,and the loading amount of the sample was 8~12 mL.The contents of glutathione,ergothiine and total polyphenol were determined.The results showed that Pleurotus ostreatus and Boletus ostreatus had higher contents of three active components,followed by Agrocybe aegerita,Cordyceps militaris and Grifola frondosa.3?The DPPH free radical scavenging rate and ABTS free radical scavenging rate of avenanthramides and mushroom extracts in vitro were tested.The results showed that the free radical scavenging capacity of avenanthramides solution after purification was greatly improved,which was 10 times of that of the result without purification,the free radical scavenging capacity at low concentration(0~20 displacement g/mL)was stronger than the Vc control at the same concentration,and the clearance rate increases with the improvement ofsolution purity,Boletus ostreatus,Cordyceps militaris,Agrocybe aegerita and Pleurotus ostreatus of the 14 kinds of mushroom showed good free radical scavenging capacity for DPPH and ABTS,probably due to their high contents of total polyphenols and ergothione;there was positive correlation between ergothione,glutathione or total polyphenol content and clearance rate,among which there was a better positive correlation between total polyphenol content and clearance rate,which was significantly higher than glutathione and ergothione.4?The results of sensory evaluation,physical and chemical properties and stability of skin care lotion were conducted.Moderate pH and viscosity of skin care lotion are suitable for skin use,in accordance with emulsion standards.The antioxidant property of skin care emulsion is good,which can effectively remove free radicals and slow down the oxidation of skin.
