Tephrosia purpurea is a source plant of rotenone.In our previous reaserchs,a number of endophytic fungi were obtained from Tephrosia purpurea.Being tested by primary screening,it was found that the strain TPL04 has potential research values.In order that compounds with activity were separated by means,the strain TPL04 was selected as the research object in this paper,from which secondary metabolites were collected by fermentation.The specific research results are as follows.?1?The strain TPL04 roots in leaves of Tephrosia purpurea.It was determinated that the strain TPL04 is Purpureocillium lilacinum by the morphological and molecular identification.After fermentation of the strain TPL04,methanol extracts from mycelium,ethyl acetate and n-butyl alcohol extracts from fermented liquid were collected,and the total amount of extracts is 48.4 g.?2?Ditylenchus destructor was used for toxic activity test.The result indicated that the mortality of nematodes reached 94.59%in 24 h,when the concentration of extracts was18.0 mg/mL.The virulence regression equation is Y=0.3147+4.9354 X?X2=1.8963?with the value of LC500 being 8.90 mg/mL.?3?Eight compounds were isolated and identified from the metabolites of the strain TPL04,they were 5-hydroxyhexan-4-olide?1?,?R?-4-benzyl-2-oxazolidinone?2?,Thymidine?3?,Methylparaben?4?,Lumichrome?5?,?-Daucosterin?6?,Uridine?7?,Hexitol?8?.Among them,compounds 1,2 and 4 were isolated from the strain TPL04 for the first time.Moreover,compound 2's configurational isomer could be obtained from metabolites of Tephrosia purpurea's leaves.Compound 4 inhibited the growth of four plant pathogens:it inhibited Pellicularia sasakii with the value of EC500 being 54.62 mg/L and Sclerotinia sclerotiorum with the value of EC500 being 179.55 mg/L in 2 d,Phytophthora capsici with the value of EC500 being 78.08 mg/L in 4 d,Botrytis cinerea with the value of EC500 being 109.32 mg/L in5 d.
