For a long time,the chemical composition,pharmacological effect and clinical research of cortex phellodendron have attracted people's attention,which resulted in the neglect of the abundant yellow pigment in the inner epidermis of cortex phellodendron.After consulting a large number of literature,scientific researchers do not report on the cortex phellodendri pigment,the study of the cortex phellodendri pigment as a natural food pigment has not been reported.Cortex phellodendron contains the natural,and more yellow pigment,yellow pigment is a very important factor in people's life is a kind of pigment,cortex phellodendron the gorgeous colour pigments that give food,safe non-toxic,for the food industry,the development of a new,natural,safe,non-toxic harmless,trust of food additive has an important significance.In this paper,cortex phellodendri pigment was taken as the research object.By comparing the three extraction methods of cortex phellodendri pigment:ultrasonic extraction,solvent extraction and microwave extraction,single factor and orthogonal experiments were carried out on the extraction process of cortex phellodendron,and the stability of cortex phellodendri pigment was also examined.Through literature investigation,it is found that the bitter taste of cortex phellodendron is caused by the presence of alkaloids in cortex phellodendron,such as berberine and obacunone in cortex phellodendron,which are characteristic substances of cortex phellodendron.Therefore,it is necessary to establish an HPLC method to simultaneously determine the berberine and obacunone in cortex phellodendron.The electronic tongue detection device was used to determine the effect of Neotame on the modification test of cortex phellodendri pigment,and the stability of Neotame was examined.Microwave extraction is the best of the three extraction methods,with the highest extraction rate and the simplest.The extraction conditions were water,the solid-liquid ratio?g:m L?was 1:100,the extraction time was 160s,and the power was 520W.Cortex phellodendri pigment is stable in light,p H?7,Na+,Cu2+,Mg2+,Ca2+ionic solutions,common additives and oxidizing substances.The linearity of berberine hydrochloride and obacunone was good in the range of 403.4-10084.7?g.The recoverises of berberine at standard concentrations of500?g,1000?g and 5000?g?n=6?were 91.27-100.2%and RSD1.58%-2.11%.The recoveries of obacunone at standard concentrations of 100?g,500?g and 1000?g?n=6?were 92-98.21%and RSD1.84-2.53%.Intraday reproducible assay RSD1.81-1.96,daytime reproducible assay RSD2.09-2.75%.The content of berberine in 3 batches was higher than that of obacunone,with the content of 14.27-15.43 mg·g and RSD 2.29-2.85%,the content of obacunone was1.15-1.34 mg·g and RSD1.15-1.56%.The color of cortex phellodendri pigment can be modified by electronic tongue test.The bitterness can be reduced and the bitterness can be improved.When the content of neotame is 0.025%,the bitterness value decreases to 1.25.When the content of neotame is increased by 0.4%,the bitterness value is 0.75?The effects of heat stability,water stability and p H on the stability of neotame were investigated.It is concluded that the thermal stability of neotame is not good above 100?,and the longer the time,the worse,so the temperature is better below 100?.Regardless of whether it was placed for 1d or 50d,the peak area of the aqueous solution of neotame did not change much,and the retention rate was greater than 94.43%.It indicates that neotame is stable in water and will not degrade or evaporate.When the p H is less than 7,the peak area has little effect.The neotame has a p H of 7?including 7?.Over time,the peak area is smaller and even undetected.It is indicated that neotame is decomposed in an alkaline solution because it accelerates the decomposition rate of neotame in an alkaline solution.It shows that neotame and cork pigment have similar stability,so neotame is very suitable for adding bitterness in the cortex phellodendri pigment to improve the taste.The best extraction method and process parameters suitable for extracting cortex phellodendri pigment were established by this study.The high-phase liquid chromatography method of berberine and obacunone in cortex phellodendron was established.The effect of neotame on the modification of cortex phellodendri pigment was analyzed by electronic tongue.The stability of neotame was determined and the study provided ideas for the subsequent development of cortex phellodendron.
