Procyanidin is a kind of bioflavonoids that own special molecular structure,which is widely found in red wine,grapes,vegetables,and other fruits.With strong antioxidative activity,it is internationally recognized as the most effective natural antioxidant used to clean up free radicals in the human body.Therefore,it has the very strong physiological activity in the body.Procyanidin B2 is one of the strongest active dimers which exists in procyanidins.Due life cycle of nematodes is short,large scale samples of life experiments can be implemented,and its genetic and environmental background related to life is clear.Therefore,the nematodes?Caenorhabditis elegans?are very suitable for evaluation of aging intervention effect and mechanism research.So this experiment was designed through the C.elegans as the study object to explore the extending lifespan effect of grape procyanidins B2.The main results are as follows:Firstly,the wild type nematodes were taken as the research object to explore the protective effect of the grape procyanidin B2 under 20?normal conditions,35?heat stress and Oxidative resistance induced by H2O2,respectively.The results showed that,compared with the control group,1?M procyanidin B2 could significantly extend the average life expectancy by 29.8%?p<0.01?in the wild type nematodes under heat stress condition,while it could prolong the average life expectancy by 28.4%?p<0.05?and 23.8%?p<0.01?of the wild type nematode under normal and oxidative stress conditions,respectively.In addition,with regard to the study of the enzyme activity of the wild type nematode,procyanidin B2 could significantly improve the activities of superoxide dismutase?SOD?and glutathione peroxidase?GSH-Px?by 41.7%and 313.6%,respecetively?p<0.01?.Furthermore,procyanidin B2 had no significant effect on the life of daf-16 mutant nematodes compared with the control group?p>0.05?.However,procyanidin B2 had significant influence on the life of wild type nematode compared with daf-16 mutant nematodes?p<0.01?.Meanwhile,procyanidin B2 was found to have no significant effect on the life of daf-2mutant nematodes compared with the control group?p>0.05?,indicating that the phenomenons of procyanidin B2 extending nematodes'lifespan are related to daf-2 and daf-16 genes.Subsequently,through the test of the fluorescence intensity in the transgenic C.elegans of daf-16::GFP and SOD-3::GFP respectively,it is found that procyanidin B2 could significantly increase the fluorescence intensity of transgenic C.elegans by19.4%and 12%,respectively?p<0.05?.Taken together,the procyanidin B2 improves the gene expressions of daf-16 and sod-3,therefore it can increase enzyme activity of SOD and GSH-Px,moreover,it enhance heat and oxidative stress abilities of C.elegans and postpone aging of the C.elegans and prolong its average lifespan.
